摘 要:目的研究肉苁蓉多糖(CDP)对D-半乳糖致衰老小鼠抗疲劳的作用及其机制。方法将80只昆明种小鼠随机分为正常对照组、模型组、CDP低剂量组和CDP高剂量组,每组20只。模型组、CDP低剂量组和CDP高剂量组予D-半乳糖100 mg/kg颈背部皮下注射造模,正常对照组予等剂量0. 9%氯化钠注射液颈背部皮下注射,连续30 d。造模同时,CDP低剂量组及CDP高剂量组分别予CDP 100、400 mg/kg灌胃,对照组和模型组予100 mg/kg蒸馏水灌胃,每日1次,连续30 d。比较4组小鼠负重游泳时间,血清尿素氮(BUN)、乳酸(LA)水平,肝糖原、肌糖原含量,肝组织超氧化物歧化酶(SOD)、丙二醛(MDA)及谷胱甘肽过氧化物酶(GSH-PX)活性。结果模型组小鼠负重游泳时间少于正常对照组(P <0. 05);与模型组相比,CDP低剂量组及CDP高剂量组小鼠负重游泳时间明显增加(P <0. 05)。模型组小鼠血清BUN及LA水平明显高于正常对照组(P <0. 05);与模型组相比,CDP低剂量组及CDP高剂量组小鼠血清BUN及LA水平明显降低(P <0. 05)。模型组肝糖原及肌糖原含量明显低于正常对照组(P <0. 05);与模型组相比,CDP低剂量组及CDP高剂量组肝糖原及肌糖原含量明显升高(P <0. 05)。与正常对照组相比,模型组小鼠肝组织SOD及GSH-PX活性降低(P <0. 05),MDA活性升高(P <0. 05);与模型组相比,CDP低剂量组及CDP高剂量组小鼠肝组织SOD及GSH-PX活性升高(P <0. 05),MDA活性降低(P <0. 05)。结论 CDP对D-半乳糖致衰老小鼠具有抗疲劳的作用,这种作用可能与降低血清BUN、LA含量,增加肝糖原、肌糖原含量,增强抗自由基损伤有关。Objective To explore the anti-fatigue effect and mechanism of cistanche deserticola polysaccharide(CDP)on D-galactose-induced aging mice.Methods 80 Kunming mice were randomly divided into normal control group,model group,CDP low-dose group and CDP high-dose group,with 20 rats in each group.The model group,CDP low-dose group and CDP high-dose group were given D-galactose 100 mg/kg for the injection of the back of the neck,and the control group was injected subcutaneously with the same dose of 0.9%sodium chloride injection for 30 days.At the same time,the CDP low-dose group and the CDP high-dose group were given CDP 100 and 400 mg/kg by gavage,respectively.the control group and the model group were given 100 mg/kg distilled water by gavage.All groups were treated once a day for 30 days.The heavy swimming time,the levels of serum urea nitrogen(BUN)and lactic acid(LA)in serum,the hepatic glycogen and muscle glycogen content,liver tissue superoxide dismutase(SOD),malondialdehyde(MDA)and glutathione peroxidase(GSH-PX)activities were compared between the four groups.Results The weight-bearing swimming time of the model group was lower than that of the control group(P<0.05).Compared with the model group,the weight-bearing swimming time was significantly increased in CDP low-dose group and CDP high-dose group(P<0.05).The serum BUN and LA levels in the model group were significantly higher than those in the control group(P<0.05).Compared with the model group,serum BUN and LA levels were significantly decreased in CDP low-dose group and CDP high-dose group(P<0.05).The hepatic glycogen and muscle glycogen content in the model group were significantly lower than those in the control group(P<0.05).Compared with the model group,the hepatic glycogen and muscle glycogen content were significantly decreased in CDP low-dose group and CDP high-dose group(P<0.05).Compared with the control group,the SOD and GSH-PX activities in the liver of the model group were decreased(P<0.05),and the MDA activity was increased(P<0.05).Compared with
