摘 要:目的优选表面活性剂-超声协同提取红景天总黄酮的工艺。方法以总黄酮提取率为评价指标,对不同类型的表面活性剂进行筛选,再分别以表面活性剂体积分数、乙醇体积分数、超声功率、提取时间、料液比进行单因素试验,采用L_(27)(3~5)正交试验优选提取工艺。结果十二烷基硫酸钠(SDS)在60%乙醇中对总黄酮的增溶效果最好;正交试验优选的最佳提取工艺为以1.5%SDS作为增溶剂,超声功率为600 W,60%乙醇为溶剂,料液比为1∶15(g∶L),提取15 min。验证试验结果显示,总黄酮的平均提取率为4.04%,RSD为0.81%(n=5)。结论所建表面活性剂-超声协同提取工艺可经济、高效地提取红景天中总黄酮。Objective To optimize the synergistic extraction process of total flavonoids from Rhodiola crenulata by surfactant-ultrasound synergistic extraction.Methods With the extraction rate of total flavonoids as the evaluation index,different types of surfactant were screened.Single factor test was carried out with the volume fraction of surfactant,the volume fraction of ethanol,ultrasonic power,extraction time and solid-liquid ratio,respectively.The extraction conditions were optimized by L27(35)orthogonal test.Results The sodium lauryl sulfate(SDS)had the best solubilization effect on flavonoids in 60%ethanol.The optimum extraction process of orthogonal test was as follows:1.5%SDS was taken as solubilizer,ultrasonic power was 600 W,60%ethanol was taken as the solvent,the solid-liquid ratio was 1∶15(g∶L),the extraction time was 15 min.The verification result showed that the average extraction rate of total flavonoids was 4.04%and RSD was 0.81%(n=5).Conclusion The established surfactant-ultrasound synergistic extraction method is economical and efficient for extraction of total flavonoids from Rhodiola crenulata.
