摘 要:目的探讨中药提取成分芍药苷对糖尿病大鼠视网膜Müller细胞的影响。方法SPF级雄性SD大鼠40只,随机分成对照组、糖尿病组、芍药苷组及二甲双胍组(阳性对照),每组10只。后3组大鼠采用单次腹腔注射链脲佐菌素(STZ)(50 mg·kg^(-1)),72 h后检测尾静脉血糖,将血糖浓度>16.7 mmol/L的大鼠作为糖尿病模型。模型诱导成功后,芍药苷组给予芍药苷(60 mg·kg^(-1))灌胃治疗,每日2次。二甲双胍组给予二甲双胍(20 mg·kg^(-1)·d^(-1))灌胃治疗。对照组、糖尿病组给予等剂量生理盐水。实验期间动态监测大鼠血糖、体重变化。12w后,试剂盒检测视网膜中谷氨酸含量变化,免疫荧光检测视网膜神经胶质酸性蛋白(GFAP)、胶质细胞谷氨酸转运体(GLAST)表达,Western blot检测视网膜GFAP、GLAST、谷氨酰胺合成酶(GS)蛋白的相对表达量。结果与对照组相比,糖尿病组血糖明显升高且均>16.7 mmol/L,GFAP表达及谷氨酸含量明显升高,GLAST、GS表达明显下降,有统计学意义(均为P<0.01);而与糖尿病组相比,芍药苷组及二甲双胍组血糖均不同程度下降,GFAP表达及谷氨酸含量明显下降,GLAST、GS表达明显增加,有统计学意义(均为P<0.01)。结论芍药苷可能通过降低大鼠血糖及抑制胶质细胞活化,上调视网膜GLAST、GS表达,降低视网膜谷氨酸含量,提示芍药苷对糖尿病大鼠视网膜Müller细胞具有保护作用。OBJECTIVE To investigate the effect of paeoniflorin on retinal Müller cells of diabetic rats.METHODS Forty SPF male SD rats were randomly divided into control group,diabetes group,paeoniflorin group and metformin group,with 10 rats in each group.Diabetes model was induced by streptozotocin(STZ,50 mg·kg^-1)in the latter three groups.After 72 hours,the blood glucose of tail vein was detected,and rats with blood glucose beyond 16.7 mmol/L were taken as models of diabetes.After the model was successfully induced,paeoniflorin group was treated with paeoniflorin(60 mg·kg^-1)by gavage,two times a day.Metformin group was given metformin(20 mg·kg^-1)per day by lavage.The control group and the diabetic group were given equal dose of normal saline.Changes of blood glucose and body weight were monitored dynamically during the experiment.After 12 weeks,the expression of retinal glial fibrillary acidic protein(GFAP)and L-glutamate/L-aspartate transporter(GLAST)and glutamine synthase(GS)were detected by immunofluorescence and Western blot.At the same time,the assay of glutamic acid in the retina by a kit.RESULTS Compared with the control group,the blood glucose level of rats in diabetic group was significantly higher than 16.7 mmol/L,the expression of GFAP and the content of glutamic acid in diabetes group significantly increased;The expression of GLAST and GS was significantly decreased(all P<0.01);Compared with the diabetic group,the blood sugar of paeoniflorin group and metformin group decreased to varying degrees,the expression of GFAP and content of glutamic acid significantly decreased,the expression of GLAST and GS in paeoniflorin group and metformin group increased significantly(all P<0.01).CONCLUSIONS Paeoniflorin may increase the expression of GLAST and GS in the retina by reducing blood sugar and inhibiting the activation of glial cells and decreasing the content of glutamic acid in the retina.The results indicated that paeoniflorin had a protective effect on diabetic rats retinal Müller cells.
