摘 要:目的观察滋肾通窍饮对实验性糖尿病(DM)大鼠血清生长激素(GH)、视网膜毛细血管基底膜厚度的影晌,探讨滋肾通窍饮对非增殖性糖尿病视网膜病变的治疗作用。方法建立链脲佐菌素(STZ)诱导的DM大鼠模型。随机分为正常组、模型组、导升明组、滋肾通窍饮低剂量和滋肾通窍饮高剂量组,放射免疫测定血清GH,电镜下观察视网膜毛细血管基底膜厚度。结果与正常组比较,STZ诱导的模型组大鼠的血糖和GH的含量显著增加,而体质量显著降低,差异具有统计学意义(P<0.05)。与模型组比较,滋肾通窍饮高和低剂量组治疗后对DM大鼠的GH具有抑制作用,差异有统计学意义(P<0.05)。与模型组比较,滋肾通窍饮高和低剂量组对DM大鼠的体质量有升高的趋势,但无统计学差异(P>0.05)。滋肾通窍饮高和低剂量组对DM大鼠的血糖具有降低的趋势,但无统计学差异(P>0.05)。与正常组比较,模型组电子密度不均匀,节段性结节样增厚。与模型组比较,滋肾通窍饮高剂量组电子密度较均匀,结构清晰,增厚不明显。结论滋肾通窍饮可降低DM大鼠的血清GH水平,并可抑制视网膜毛细血管基底膜增厚,对视网膜毛细血管起到保护作用。OBJECTIVE To observe the effect of Zishen Tongguan yin(ZSTQY)on serum growth hormone,retinal capillary basement membrane thickness in experimental diabetes mellitus(DM)rats,and to explore the effect of ZSTQY on non-proliferative diabetic retinopathy.METHODS The streptozotocin(STZ)-induced DM rats were randomly divided into normal group,model group,doxium group,ZSTQY low dose and ZSTQY high dose group.We used radioimmunoassay to determine the serum growth hormone,and electron microscope was used to observe retinal capillary basement membrane thickness.RESULTS Compared with the normal group,the content of blood glucose and growth hormone STZ-induced model rats increased significantly,while the body weight was significantly decreased,the difference was statistically significant(P<0.05).Compared with the model group,high and low dose ZSTQY group showed significant inhibitory effect on growth hormone in DM rats,the difference was statistically significant(P<0.05).Compared with the model group,the body weight of DM rats increased with the high and low dosage of ZSTQY,but there was no significant difference(P >0.05).In addition,the blood glucose level of high dose group and low dose group both decreased,but there was no significant difference between the two groups(P >0.05).Compared with the normal group,the electron density of the model group was uneven,and nodular thickening was observed.Compared with the model group,the high dose ZSTQY group showed uniform electron density,clear structure and no thickening change.CONCLUSIONS The ZSTQY reduced serum level of growth hormone in DM rats,and inhibited the thickening of retinal capillary basement membrane,thus it was of protective effect on retinal capillary layer.
