摘  要:为了对鸡集约化养殖场的疑似鸡毒支原体病死鸡样品进行检测,基于crmA基因和lipoprotein基因通过PCR方法进行分子鉴定,并对脂蛋白(Lipoprotein,LP)进行生物信息学分析,探索该蛋白的结构并预测其功能,为其蛋白功能的研究提供一定的理论基础。通过PCR方法获得了预期片段并进行测序分析,通过生物信息学软件对脂蛋白进行预测分析。结果显示,经测序鉴定为鸡毒支原体,预测LP蛋白总体亲水性较低,为疏水蛋白,二级结构包括α螺旋、β转角、无规则卷曲和延伸链,并具有5个潜在的优势抗原表位,可作为一种候选疫苗抗原。表明对本地区分离株病原进行分子鉴定研究,对诊断预防该病的发生都具有重要的现实意义;提示控制此病的发生流行,坚持预防为主,加强饲养管理,积极防治鸡毒支原体感染的发生。This study was conducted to detect the pathogen which caused chicken death and analyze the lipoprotein(LP)to predict its function.The molecular dection and identification of Mycoplasma gallisepticum from the suspected dead chicken samples from intensive chicken farm was carried out using polymerase chain reaction(PCR)based on crmA genes and lipoprotein gene,The lipoprotein(LP)was analyzed by bioinformatics methods,and the structure and the function of the protein was explored and predicted,The pathogen was identified as M.gallisepticum by polymerase chain reaction(PCR)to obtain the expected gene fragments and sequencing analysis.By using the bioinformatics software,the lipoprotein was identified to be a hydrophobic protein with low hydrophilicity,which contains alpha helix(α-helix),beta angle,random coil and chain extension,and have five preponderant potential antigen epitopes,it can be used as a vaccine candidate.Therefore,it was necessary to isolate the pathogenic strains and do molecular identification,also was important and significant to diagnose and prevent the disease occurring;it prompted that put prevention first,strengthened breeding managements,and controled and treated M.gallisepticum infection occurring actively.
