白细胞介素-17A及白细胞介素-17F基因多态性与中国汉族人群强直性脊柱炎的相关性分析

摘　　要：目的探讨中国汉族人群中白细胞介素(IL)-17A基因多态性位点rs2275913及IL-17F基因多态性位点rs763780与强直性脊柱炎(AS)发病的相关性。方法选取汉族人群AS患者46例和同期健康体检者33名,采用MassArray基因分型方法检测IL-17基因多态位点,计算等位基因及基因型频率分布,并进行统计学分析。结果 IL-17A基因rs2275913位点等位基因频率在2组间分布差异有统计学意义(P=0.03),携带IL-17基因rs2275913位点A等位基因与G等位基因比较,显著增加AS的罹患风险[OR (95%CI)=2.02 (1.08,3.79)];而IL-17F基因rs763780位点的基因型及等位基因频率分布在AS组和对照组间差异无统计学意义(P>0.05)。其各个基因型组的红细胞沉降率和C反应蛋白水平差异无统计学意义(P>0.05)。结论在中国汉族人群中,IL-17A基因rs2275913多态性与AS的易感性相关,其中携带IL-17A rs2275913的A基因增加AS的罹患风险;IL-17F基因rs763780位点多态性与AS的易感性无关。Objective To investigate the genetic association of interleukin(IL)-17 polymorphisms with ankylosing spondylitis(AS)in Han Chinese population.Methods The study group consisted of 46 patients with AS and of 30 healthy individuals.SNPs in the IL-17A(rs2275913)and IL-17F(rs763780)genes were genotyped using Mass Array assays.Allele and genotype frequencies were calculated and statistical analyzed.Results The distribution difference in the allele frequency of rs2275913 between AS group and the control group was statistically significant(P=0.03),carrying A genetic mode of IL-17A rs2275913 increases significan the risk of AS compared with G[OR(95%CI)=2.02(1.08,3.79)].The allele and genotype frequencies of rs763780 in IL-17F gene were not different between patient with AS and control.No significant associations were observed with erythrocyte sedimentation rate(ESR)and C reactive protein(CRP)among all gene groups(P >0.05).Conclusion Polymorphism of IL-17A gene rs2275913 is related to susceptibility of AS in Han Chinese population.A gene carrying IL-17A rs2275913 increases the risk of AS.There was no significant correlation between SNPs of IL-17F gene rs763780 and susceptibility to AS.
