加味大柴胡汤对急性胰腺炎大鼠的炎症抑制作用

摘　　要：目的观察加味大柴胡汤对急性胰腺炎(AP)大鼠模型胰腺病理组织和对白细胞介素-6(IL-6)、白细胞介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)等炎症因子的影响并探讨其机制。方法采用随机数字表法将实验大鼠分为假手术组、AP组、治疗组,治疗组给予加味大柴胡汤灌胃;AP组和假手术组予等量0.9%氯化钠注射液灌胃,治疗1周后对大鼠进行造模,并于24 h后处死实验大鼠,并快速切取其胰腺组织,采用Kusske法进行病理评分;另从心脏取血,采取ELISA法检测IL-6、IL-8和TNF-α水平,并进行统计学分析。结果 AP组及治疗组的胰腺组织炎症程度均较假手术组严重,炎症因子水平亦较假手术组升高;而治疗组胰腺炎症程度较AP组明显减轻;治疗组的炎症因子水平较AP组均有明显下降(均P <0.05)。结论加味大柴胡汤在改善大鼠的胰腺组织炎性改变,抑制IL-6、IL-8、TNF-α等炎症因子方面的作用是确切的,这也为其临床的应用和推广提供了实验依据。Objective:To observe the effect of Modified Dachaihu Decoction on pancreatic pathology and inflammatory factors,such as IL-6,IL-8 and TNF-αin rats with acute pancreatitis and explore its mechanism.Methods:The rats were divided into three groups by random number table:sham operation group,AP group and the treatment group.The treatment group was given Modified Dachaihu Decoction by gavage,and AP group and sham operation group were given the same amount of saline by gavage,once every 12 hours.After one week′s treatment,rats were modeled and killed 24 hours later.The pancreas tissues were cut quickly and the pathological scores were performed with Kusske method,and the levels of IL-6,IL-8 and TNF-αwere detected by ELISA from the heart.Results:The severity of pancreatic inflammation in AP group and the treatment group was more serious than that in sham operation group,and the level of inflammatory factors was also higher than that in sham operation group.The degree of pancreatic inflammation in the treatment group was significantly less than that in AP group,and the level of inflammatory factors in the treatment group was significantly lower than that in AP group(P<0.05).Conclusion:Modified Dachaihu Decoction can improve the inflammatory changes of pancreatic tissue and inhibit inflammatory factors,such as IL-6,IL-8 and TNF-αin rats,providing experimental basis for its clinical application and promotion.
