健脾益气通络中药对糖尿病肾病大鼠肾组织病理变化的影响

摘　　要：目的观察健脾益气通络中药对糖尿病肾病大鼠肾组织病理变化的影响。方法将实验用的60只健康雄性SD大鼠,随机法分为6组:左肾切除组、模型组、洛汀新组、中药低、中、高剂量组,每组收入10只,除肾切除组外,其余五组均构建糖尿病肾病大鼠模型,并对中药低、中、高剂量组进行对应剂量的健脾益气通络中药灌胃,洛汀新组进行洛汀新灌胃,对左肾切除组、模型组进行等量生理盐水灌胃,灌胃6周后,检测各组大鼠24h的尿蛋白(Upro)定量、肌酐(Scr)、尿素氮(BUN)。取肾脏开展HE染色行电镜观察以及透射电镜观察肾皮质组织病理改变。结果模型组大鼠与肾切组大鼠相比,光镜及电镜下已出现糖尿病肾病病理改变,各治疗组肾皮质病理改变较轻;洛汀新组、中药低、中、高剂量组与模型组比较,24h尿蛋白(Upro)定量显著下降(P <0. 01);各治疗组与模型组比较,尿素氮(BUN)、肌酐(Scr)明显降低(P <0. 05)。结论健脾益气通络中药可改善糖尿病肾病大鼠的肾功能,并可减轻肾脏的病理损害。Objective:To observe the effect of Jianpi Yiqi Tongluo Recipe on the pathological changes of renal tissue in rats with diabetic nephropathy.Methods:60 healthy male SD rats were randomly divided into 6 groups:left nephrectomy group,model group,lotensin group,low,medium and high dose TCM groups,10 in each group.Except the nephrectomy group,the other five groups were established as rat models of diabetic nephropathy.The low,medium and high dose TCM groups were given intragastric administration of corresponding dose of Jianpi Yiqi Tongluo,and lotensin group was were intragastrically administrated with lotensin.The rats in the left nephrectomy group and the model group were intragastrically administered with normal saline.After 6 weeks of gavage,the urine protein(Upro),creatinine(Scr)and urea nitrogen(BUN)were measured in each group for 24 hours.The kidneys were subjected to HE staining by electron microscopy and transmission electron microscopy to observe the pathological changes of renal cortex tissue.Results:Compared with the nephrectomized rats,there was the pathological changes of diabetic nephropathy in the model group under electron microscopy and transmission electron microscopy and there was lighter pathological changes of renal cortex in each treatment group.Compared with the model group,the 24h urine protein(Upro)ration of the lotensin group and the low,medium and high dose TCM groups was significantly decreased(P<0.01).Compared with the model group,the urea nitrogen(BUN)and creatinine(Scr)in each treatment group were significantly lower than those in the model group(P<0.05).Conclusion:Jianpi Yiqi Tongluo Recipe can improve the renal function of rats with diabetic nephropathy and reduce the pathological damage of the kidney.
