摘 要:目的探讨黄连素在大鼠肾脏缺血再灌注损伤中的作用。方法 40只雄性SD大鼠随机分为对照组(S组)、模型组(IR组)、黄连素组(H组)和黄连素+Exendin-(9-39)组(HE组)。S组给予生理盐水及行假手术处理,IR组给予生理盐水,H组给予黄连素,HE组给予黄连素+Exendin-(9-39),三组制作大鼠肾脏缺血再灌注模型。观察肾组织病理学改变,检测造模前及造模12 h和24 h后血清中肌酐(SCr)、尿素氮(BUN)水平及血清白细胞介素(IL)-10、血管内皮生长因子(VEGF)以及肾组织中肿瘤坏死因子(TNF)-α、IL-1β、IL-6的含量。本次实验研究时间为2016年9~11月。结果模型制作成功,血清BUN、Cr水平在H组中低于IR组(P<0.05),而HE组低于H组(P<0.05);IR组、H组、HE组中IL-10、IL-6、TNF-α、VEGF水平均高于S组(P<0.05),HE组IL-6、TNF-α水平较H组明显降低(P<0.05)。四组IL-1β水平无明显变化。结论黄连素能有效改善肾脏缺血再灌注大鼠的肾功能,其机制可能与抑制肾脏组织炎症反应有关。Objective To investigate the role of berberine in renal ischemia-reperfusion injury in rats.Methods 40 male Sprague-Dawley rats were randomly divided into control group(group S),model group(group IR),berberine group(group H)and berberine+Exendin-(9-39)group(group HE).Group S was given saline and sham surgery;group IR was given saline,group H was given berberine,and group HE was given berberine+Exendin-(9-39).Rat kidney ischemia-reperfusion models were made in the three groups.The pathological changes in renal tissues were observed.Serum creatinine(SCr),urea nitrogen(BUN)levels,and serum interleukin(IL)-10,vascular endothelial growth factor(VEGF),tumor necrosis factor in kidney tissues,as well as the contents of(TNF)-α,IL-1β,and IL-6 were measured before modeling and 12 h and 24 h after modeling.The study time of his experiment was from September 2016 to November 2016.Results The model was successfully established.Serum BUN and Cr levels were lower in the group H than in the group IR(P<0.05).The group HE was lower than the group H(P<0.05);the levels of IL-10,IL-6,TNF-αand VEGF in group IR,group H and group HE were all higher than those in group S(P<0.05).The levels of IL-6 and TNF-αin the group HE were significantly lower than those in the group H(P<0.05).There were no significant changes in the levels of IL-1βin the four groups.Conclusion Berberine can effectively improve the renal function of the rats receiving renal ischemia-reperfusion.Its mechanism may be related to the inhibition of inflammatory reactions in the kidney tissues.
