摘  要:目的分析2017年宜昌市流感疫情中BY流感毒株的HA和NA基因特性,了解其与WHO推荐的系参考株、疫苗株和当地代表株的匹配情况。方法对流感疫情病例标本中分离得到的7株BY型流感毒株的HA和NA进行扩增和序列测定,通过Mega 5.0软件对测序产物进行分析。结果 7例疫情毒株均为BY系的HA蛋白的3分枝,相互间HA基因的核苷酸和氨基酸同源性达98.9%和99.3%;与WHO疫苗株B/PHUKET/3073/2013比较,进化距离接近,没有发现氨基酸序列的丢失与插入,HA基因有2个氨基酸位点发生变化,NA基因有11个氨基酸位点发生变化。结论引起流感疫情的毒株目前变异不明显,与WHO疫苗株匹配,应加强重点人群的疫苗接种。开展连续流感分子生物学监测,及时分析病毒基因特性,对流感的防控有重要意义。Objective To analyze the genetic characteristics of the HA and NA genes of influenza B virus isolated from successive influenza outbreaks in Yichang in 2017,and to match it with the WHO reference strain,vaccine strain and local representative strain.Methods The HA and NA genes of 7 influenza B viruses isolated from specimens were amplified and sequenced,and the sequencing products were analyzed by the software Mega 5.0.Results All the 7 strains belonged to the third branch of the BY family of HA proteins,and the homology of the nucleotide and amino acid was 98.9%and 99.3%,respectively.Compared with the WHO vaccine strain(B/PHUKET/3073/2013),the evolutionary distance was close,and the loss and insertion of amino acid sequences were not found.There were 2 variations in the amino acid sequence in HA and 11 variations in the amino acid sequence in NA.Conclusion The virus causing influenza outbreak does not have obvious mutations.Matching with WHO vaccine strains,vaccination of key populations should be strengthened.It is important to carry out continuous influenza molecular biological monitoring and timely analysis of virus gene characteristics for the prevention and control of influenza.
