摘 要:目的观察中药滋肾清心汤对卵巢储备下降(DOR)小鼠的影响,并探讨滋肾清心汤可能的作用机制。方法雌性BALB/c小鼠随机分为4组:对照组、DOR组、滋肾清心汤+DOR组(中药组)和脱氢表雄酮+DOR组(西药组),每组10只。比较各组小鼠卵巢中卵泡计数及Caspase-3表达情况,并采用ELISA试剂盒检测小鼠血清中雌二醇、卵泡刺激素和抗苗勒管激素水平。结果滋肾清心汤和脱氢表雄酮治疗均可改善DOR组小鼠窦前卵泡、窦卵泡和闭锁卵泡变化。滋肾清心汤和脱氢表雄酮治疗均可减少DOR组小鼠卵巢细胞Caspase-3表达。此外,DOR组小鼠血清雌二醇、卵泡刺激素和抗苗勒管激素水平均降低;而滋肾清心汤和脱氢表雄酮治疗均可提高激素水平。结论滋肾清心汤可能通过调节Caspase-3表达改善DOR小鼠卵巢储备。Objective To observe the effect of zisheqingxin decoction on diminished ovarian reserve(DOR)mice,and explore the possible mechanism.Methods Female BALB/c mice were randomly divided into 4 groups:control group,DOR group,zisheqingxin decoction+DOR group(Chinese medicine group)and dehydroepiandrosterone+DOR group(Western medicine group),each group had 10 mice.The follicle count and apoptosis in ovaries were compared;ELISA reagent was used to measure the levels of estradiol,follicle stimulating hormone and anti-Mullerian hormone level in mice.Results The treatment of zisheqingxin decoction and dehydroepiandrosterone could improve the changes of preantral follicles,sinus oocytes and atresia follicles in DOR group.The treatment of zisheqingxin decoction and dehydroepiandrosterone could reduce the Caspase-3 of mouse ovarian cells in DOR group.In addition,the level of serum estradiol,follicle stimulating hormone and anti Mullerian hormone were all decreased in DOR group,the zisheqingxin decoction and dehydroepiandrosterone treatment could relieve hormone levels.Conclusion Zisheqingxin decoction might improve the ovarian reserve of DOR mice by regulating caspase-3 expression.
