摘 要:目的研究白龙灵沙汤对小鼠Lewis肺癌生长的抑制作用。方法将40只荷Lewis肺癌C57BL/6雄性小鼠随机分为模型组及白龙灵沙汤高、中、低剂量组,每组10只。取传代好的带瘤小鼠的实体瘤块,研磨后右腋下皮下接种。接种后次日,白龙灵沙汤高、中、低剂量组分别给予33. 33 g/(kg·d)、16. 67 g/(kg·d)、8. 33 g/(kg·d)白龙灵沙汤灌胃,模型组给予等体积生理盐水灌胃,均1次/d,连续28 d。期间观察各组小鼠的一般状况,末次给药24 h后取瘤块称质量,计算肿瘤抑制率,采用流式细胞仪检测移植瘤细胞周期和细胞凋亡率,采用免疫组化方法检测肿瘤组织中微血管密度。结果白龙灵沙汤高剂量组小鼠的一般状况明显好于模型组。白龙灵沙汤各剂量组小鼠瘤质量均明显低于模型组(P均<0. 05),且白龙灵沙汤高、中剂量组均明显低于白龙灵沙汤低剂量组(P均<0. 05);白龙灵沙汤高、中剂量组抑瘤率均明显高于白龙灵沙汤低剂量组(P均<0. 05);白龙灵沙汤高、中剂量组G0/G1期细胞比例和细胞凋亡率均明显高于模型组和白龙灵沙汤低剂量组(P均<0. 05),S期细胞比例明显低于模型组和白龙灵沙汤低剂量组(P均<0. 05);白龙灵沙汤各剂量组小鼠肿瘤组织中微血管密度均明显低于模型组(P均<0. 05),且白龙灵沙汤高、中剂量组均明显低于白龙灵沙汤低剂量组(P均<0. 05)。结论白龙灵沙汤能通过抑制细胞周期促进细胞凋亡,从而降低小鼠Lewis肺癌组织中微血管密度,具有明显抑瘤作用。Objective It is to study the inhibitory effect of Bailonglingsha Decoction on the growth of Lewis lung cancer in mice.Methods Forty male Lewis lung cancer C57BL/6 mice were randomly divided into model group and Bailonglingsha Decoction groups of high,medium and low dose,with 10 rats in each group.The solid tumor blocks from the tumor-bearing mice were grinded and inoculated subcutaneously under the right axilla.On the day after inoculation,Bailonglingsha Decoction groups of high,medium and low dose were given 33.33 g/(kg·d),16.67 g/(kg·d)and 8.33 g/(kg·d)Bailonglingsha Decoction respectively by gavage.The rats in the model group were given an equal volume of normal saline.All 1 time/d,for 28 days.The general condition of the mice in each group was observed.The mass of the tumor was weighed in 24 hours after the last administration,the tumor inhibition rate was calculated,the cell cycle and apoptosis rate of the transplanted tumor were detected by flow cytometry,and microvascular density in the tumor tissue was detected by immunohistochemistry.Results The general condition of the high dose group of Bailonglingsha Decoction group was significantly better than that of the model group.The tumor quality of every Bailonglingsha Decoction group was significantly lower than that of the model group(P<0.05),and that in the high and middle dose groups were significantly lower than that in the low dose group(P<0.05);the inhibition rate of high and middle dose groups was significantly higher than that of the low dose group(P<0.05);The proportion of G 0/G 1 cells and apoptosis rate in the high and middle dose groups were significantly higher while the proportion of cells in S phase was significantly lower than those of model group and low dose group(P<0.05).The microvessel density in the tumor tissues of every Bailonglingsha Decoction group was significantly lower than that in the model group(P<0.05),and the density in the high and middle dose groups were significantly lower than the low dose group(P<0.05).Conclusion Bailon
