摘 要:目的优化益阴消渴方分离纯化工艺。方法采用大孔树脂柱层析法,以高效液相色谱法测定葛根素含量,考察乙醇洗脱用量、大孔树脂用量、树脂径高比、药液上样浓度、药液pH对富集纯化工艺的影响。结果经验证试验表明,富集纯化后益阴消渴方中葛根素的含量比上柱前葛根素的含量显著提高,葛根素转移率达85. 14%。结论益阴消渴方经大孔树脂富集纯化后,以葛根素为代表的有效部位群含量显著提高,工艺条件稳定可行,为制剂成型工艺提供了理论依据。Objective To optimize the separation and purification technology of Yiyin Xiaoke Dropping Pills.Methods Macroporous resin column chromatography method was adopted,HPLC method was used to determine the content of puerarin.The effects of ethanol elution amount,the dosage of macroporous resins,the ratio of diameter-to-height on resins,the sample concentration and pH of liquid on the enrichment and purification technology were investigated.Results After enrichment and purification,the experimental results showed that the content of puerarin in Yiyin Xiaoke recipe was significantly higher than that before using column,and the transfer rate of puerarin was 85.14%.Conclusion After enrichment and purification with macroporous resin,the content of effective site group which was represented by puerarin in Yiyin Xiaoke recipe is significantly increased,and the technological conditions are stable and feasible,which provides the theoretical basis for the preparations technology.
