摘 要:目的建立大鼠3期压力性损伤模型,并探讨活性氧在损伤过程中的作用机制。方法 60只SD大鼠随机分为正常组、假手术组,和实验组3组。试验组根据缺血再灌注损伤理论,采用臀肌下埋植铁片,外置磁铁循环压迫法,建立3期压力性损伤模型;假手术组只埋植铁片,不做压迫;正常组不做任何处理。成模后HE染色对比3组病理表现、活性氧平均荧光强度,血清超氧化物歧化酶活力和丙二醛含量。结果试验组血清SOD含量显著低于正常组和假手术组(P<0.01),MDA含量和活性氧平均荧光强度显著高于上述两组(P<0.01)。结论缺血再灌注损伤产生的活性氧,是大鼠3期压力性损伤过程中的重要因素。Objective To establish a pressure injury model of stage 3 by ischemia-reperfusion theory, and investigate the mechanism of reactive oxygen species (ROS) during the injury. Methods Sixty SD rats were randomly divided into 3 groups: Normal group, sham operation group, and experimental group. The experimental group used a circular iron piece implanted under the muscle of the buttocks and a magnet compressed on the skin outside to establish a pressure injury model of stage 3. While the sham operation group only implants iron pieces but without magnet compressing, and the normal group had no treatment. After the model was established, the HE staining pathological, ROS fluorescence intensity , serum superoxide dismutase (SOD) activity and malondialdehyde (MDA) content were compared between the 3 groups. Results Compared with the sham operation group and normal group, the SOD content of the experimental group was significantly lower (P<0.01), while the MDA content was significantly higher (P<0.01). The ROS fluorescence intensity was significantly increased in the experimental group (P<0.01). Conclusion ROS plays an important role in the process of stage 3 pressure injury of rats.
