摘  要:优化单侧肾切除联合链脲佐菌素(STZ)诱导的糖尿病肾病(DKD)大鼠建模方法,研究该模型大鼠的肾脏病变特点,为药物药效学评价和临床基础研究提供稳定平台。选取健康雄性大鼠12只,随机分为单侧肾切组(UNX组)和糖尿病肾病组(DKD组)。采用背侧横行切口配合皮内缝合的方法切除右肾,术后大鼠腹腔一次性注射STZ 40mg/kg诱导糖尿病,UNX组大鼠给予相同剂量柠檬酸缓冲液。动态检测两组大鼠体重、血糖、24h尿蛋白变化,并于第12周末观察大鼠肾脏病变。采用该手术方式,大鼠术后成活率可达100%。与UNX组相比,DKD组大鼠体重增长缓慢或负增长(P<0.01),24h尿蛋白明显增多(P<0.01),血糖维持在28mmol/L^32mmol/L。肾脏病变符合早期DKD表现。背侧横行切口配合皮内缝合右肾摘除术,联合腹腔注射STZ诱导的DKD大鼠造模方法成模率高。该模型与人类糖尿病肾病早期临床症状、病理变化相似,适用于DKD的机制研究及药物研发。To provide stable platform for the study of pharmacodynamic evaluation and clinical basic research,we optimized the method of diabetic kidney disease animal model in uninephrectomized streptozotocin-induced diabetic rats and studied the characteristics of renal lesions in the model.A total of 12 male Wistar rats were randomly divided into UNX group and DKD group.The right nephrectomy was made by dorsal lateral cut and intradermal suture.DKD model was made by administration of streptozotocin(STZ) in a single dose of 40 mg/kg intraperitoneally after operation.The rats in UNX group were given the same dose of citric acid buffer.The body weight,blood glucose and 24 h urine protein were tested.And the pathology of the kidney was observed at the end of 12 weeks.The survival rate of the rats was 100% after operation.The body weight of rats in the DKD group decreased significantly compared with that in the UNX group ( P <0.01).And the 24-h urine protein was significantly increased compared with that in the UNX group ( P <0.01).The random blood sugar was maintained between 28 mmol/L-32 mmol/L.The animal model established by dorsal lateral nephrectomy and STZ injection provided an effective method,which is in accordance with human early DKD manifestation.It is suitable for studies on drug pharmacodynamic evaluation and basic clinical studies.
