摘 要:目的探讨不同浓度脂肪干细胞对自体脂肪移植的作用,为临床提供理论依据。方法 BALB/c裸鼠随机分为4组(n=2),裸鼠背部两侧筋膜下分别注入0.5 mL待移植脂肪颗粒与不同浓度脂肪干细胞的混合物,脂肪干细胞浓度如下:200μL脂肪颗粒.A组1×10~6、B组1×10~5、C组1×10~4脂肪干细胞,D组(对照组)注入单纯脂肪颗粒。每2周测量脂肪团块长短径观察裸鼠背部脂肪团块生长情况。6周后处死裸鼠,取出脂肪组织,称质量,H-E染色及CD31血管免疫组化染色对脂肪组织存活、脂肪细胞形态及组织血管化状态进行评价。结果脂肪团块长短径测量显示,脂肪团块均有缩小且各组间差异无统计学意义。6周后,A、B、C、D各组之间脂肪组织质量差异均有统计学意义(P<0.01)。H-E染色提示B、C、D组脂肪细胞生长良好。CD31血管免疫组化检测显示B组微血管形成情况良好。结论脂肪干细胞浓度过高时,与脂肪组织的生长竞争导致脂肪细胞的存活率降低。受区空间容量也对脂肪干细胞混合脂肪颗粒的生长产生了影响。Objective To investigate the effect of different concentrations of human adipose-derived stem cells(ADSCs)on fat transplantation in nude mice.Methods Human ADSCs were isolated and cultured.Human fat granules were transplanted into fascia of the back of BALB/c nude mice mixed with 1×10^6(group A),1×10^5(group B),1×10^4(group C)ADSCs per 100μL fat granules;or fat granules were transplanted without ADSCs(group D),with 2 mice in each group.The growth of adipose mass in the back of nude mice was measured.After 6 weeks,the nude mice were sacrificed,the adipose tissue was removed and weighed;the H-E staining and the CD31-immunohistochemistry of adipose tissue were performed.The survival of adipose tissue,adipocyte morphology and vascularization were evaluated.Results During the observation period of 6 weeks,there were no significant differences in the size of fat mass between the groups.After 6 weeks,there were significant differences in wet weight of fat mass among groups A,B,C and D(P<0.01).H-E staining showed that adipocytes in group B,C and D grew well.CD31 immunohistochemical staining showed that microvessel formation in group B was good.Conclusion The high concentration of adipose-derived stem cells would affect the survival of adipocytes,and the growth of transplanted fat tissue.
