夏枯草醇提物抑制肿瘤细胞的研究

摘　　要：目的采用水提醇沉法将夏枯草成分分成醇溶出部分和醇沉淀部分,并就这两部分药物对肺癌A549细胞和人肝癌HepG_2细胞的增殖、迁移作用进行研究。方法采用80%以上的乙醇浓度对夏枯草水煎液进行醇沉法提取,不同生药浓度的夏枯草醇溶物和醇沉物处理A549细胞和HepG_2细胞,用倒置显微镜观察细胞形态的变化,MTT法检测药物对肿瘤细胞增殖影响,细胞划痕实验检测对细胞迁移的影响。结果 MTT检测发现醇沉淀物各浓度抗肿瘤效果呈浓度依赖性,浓度越高,抑制肿瘤细胞增殖的效果越强,浓度<0.25g/ml后基本无抑制作用。醇溶解物组在0.05～1.0g/ml浓度范围内呈"V"字型的抑制作用,在浓度为0.1g/ml时抑制作用最强,<0.05g/ml后基本无抑制作用。细胞形态观察与MTT结果一致,抑制作用越强,细胞圆缩,状态较差,数量少。细胞划痕实验结果发现醇溶物与醇沉物抑制细胞迁移效果呈浓度依赖性,浓度高,抑制迁移效果越强,该结果与MTT检测出现中间某浓度抑制细胞增殖效果最强的结果不一致,且醇溶物对HepG_2细胞的抑制迁移效果较A549更强。结论夏枯草各种提取物均可抑制A549细胞和HepG_2细胞的增殖和迁移,醇溶物MTT结果显示在一定范围内对A549细胞和HepG_2细胞的增殖呈"V"字型的抑制作用。Objective To study the effect of the components of Prunella vulgaris L divided into alcohol-soluble parts and the alcohol precipitation parts by water-extraction and alcohol-precipitation method on growth,migration in lung cancer A549 cells and human hepatocellular carcinoma HepG 2 cells.Methods Prunella vulgaris L was extracted with water,then added alcohol to 80%to acquire the alcohol-soluble parts and the alcohol precipitation parts.The A549 cells and HepG 2 cells were treated by different concentrations of drugs.MTT and scratch test were used to detect the growth and migration inhibitory effect on A549 cells and HepG 2 cells.Results The alcohol precipitation parts of Prunella vulgaris L could inhibit A549 cells and HepG 2 cells growth and migration,and the inhibition depended on the dosage of the drugs.When the concentration was less than 0.25g/ml,there was no inhibition.The alcohol-soluble parts of Prunella vulgaris L also could inhibit both types of cells,but the trend of inhibition presented"V"type when the concentration range was from 0.05-1.0g/ml.The inhibition effect was the strongest when the concentration was 0.1g/ml.The results of the cell morphology observation was consistent with the MTT results.The stronger the inhibitory effect,the smaller the cell size,the poorer state and the smaller number of cells.Cell scratch experiment found that the alcohol-soluble parts and the alcohol precipitation parts inhibited cell migration effect was concentration dependent.The higher the concentration,the stronger the effect of inhibiting the migration.The inhibitory effect of alcohol solution on HepG 2 cells was stronger than that of A549.Conclusion The extracts from Prunella vulgaris L inhibited the proliferation and migration of A549 cells and HepG 2 cells.
