摘  要:目的初步探讨Toll样受体(toll like receptors,TLR)及胸腺基质淋巴细胞生成素(thymic stromallymphopoietin,TSLP)在特应性皮炎(atopic dermatitis,AD)发病机制中的作用。方法将32只6~8周SPF级雌性纯系BALB/c小鼠按照随机数字表法分为对照组和实验组,每组16只。建立小鼠特应性皮炎模型,采用免疫组织化学方法检测小鼠皮损中TLR2、TSLP的表达。ELISA法测定血清中TLR2、TSLP、IL-4、IFN-γ表达水平。结果 AD小鼠皮损中TLR2的阳性表达物质主要位于细胞膜和胞浆,基底层及棘细胞层广泛表达。实验组平均光密度值(136.591±15.621)明显高于对照组(13.542±3.865)。TSLP阳性表达物质主要位于胞核,胞浆及胞膜少量着色,实验组平均光密度值(74.306±17.898)明显高于对照组(46.093±3.347)。AD小鼠血清TLR2(142.896±11.934)、TSLP(213.621±34.666)、IL-4(97.831±15.882)、IFN-γ(70.999±9.604)的表达水平明显高于对照组(14.302±3.548)、(151.997±22.328)、(60.560±9.286)、(51.224±4.534),TLR2与TSLP、IL-4表达水平呈正相关(r=0.854,0.997,P<0.01);但与IFN-γ表达水平呈负相关(r=-0.756,P<0.05)。结论 TLR2、TSLP在AD小鼠血清及皮损中大量表达,上皮细胞分泌的TLR2可通过TSLP,连接上皮细胞介导的固有免疫和适应性免疫。Objective To explore the role of toll-like receptors(TLR)and thymic stromal lymphopoietin(TSLP)in the pathogenesis of atopic dermatitis(AD).Methods A total of 32 SPF-grade female BALB/c mice were randomly divided into the control group and experiment group with 16 mice in each group.The mouse model with AD was established.The expression of TLR2 and TSLP in the lesions was tested by the immunohistchemical technique and the expression levels of serum TLR2,TSLP,IL-4 and IFN-γ were detected by enzyme-linked immunosorbent assay(ELISA).Results The positive-expression substance of TLR2 in the skin lesions was mainly located in the cell membrane and cytoplasm,the basal layer and the acanthosis layer were widely expressed,and the average light density of the experiment group(136.591±15.621)was significantly higher than that of the control group(13.542±3.865).The positive-expression substance of TSLP was mainly located in the nucleus,the cytoplasm and cell membrane was a little colored,and the average light density of the experiment group(74.306±17.898)was significantly higher than that of the control group(46.093±3.347).The expression levels of serum TLR2,TSLP,IL-4 and IFN-γ of the experiment group were(142.896±11.934),(213.621±34.666),(97.831±15.882),and(70.999±9.604)respectively,and they were significantly higher than those of the control group which were(14.302±3.548),(151.997±22.328),(60.560±9.286)and(51.224±4.534)respectively.The expression of TLR2 was positively correlated with the expression of TSLP and IL-4 and negatively correlated with the expression of IFN-γ(P<0.05).Conclusion TLR2 and TSLP are abundantly expressed in the serum and skin lesions of AD mice.TLR2 secreted by epithelial cells can connect the innate and adaptive immunity mediated by epithelial cells through TSLP.
