海南黎族、汉族人群VEGFA基因SNP与血浆VEGFA表达水平的相关性分析

摘　　要：目的:分析海南黎族、汉族血管内皮生长因子A(VEGFA)基因单核苷酸多态性(SNP)和血浆VEGFA表达水平之间的关系,探讨VEGFA基因SNP对慢性阻塞性肺疾病(COPD)诊断的意义。方法:2014年7月至2015年3月,选取海南COPD患者249例,其中黎族109例,汉族140例;对照组246例,其中黎族89例,汉族157例。通过对VEGFA基因的9个SNP位点进行分型,测定VEGFA的表达水平,分析VEGFA基因SNP与血浆VEGFA表达水平的相关性。结果:VEGFA基因rs833068,rs3024994,rs3024997和rs3025000四个位点的等位基因在汉、黎两族中分布规律有所不同。rs3025030和rs3025033位点与汉族COPD发病明显相关。rs3025030及rs3025033可增加血浆中VEGFA表达水平。结论:VEGFA基因的SNP对汉族、黎族群体血浆VEGFA表达水平的影响存在差异。Objective: To analyze single nucleotide polymorphism (SNP) of vascular endothelial growth factor A (VEGFA) gene and the plasma level of VEGFA, and to explore the application of SNP of VEGFA in diagnosis of chronic obstructive pulmonary disease (COPD). Methods: A total of 249 COPD patients from Hainan Province were selected from July 2014 to March 2015, including 109 cases of Li nationality and 140 cases of Han nationality. Besides, 246 healthy persons were selected as control group, including 89 cases of Li nationality and 157 cases of Han nationality. Genotyping of 9 SNP loci of VEGFA were performed, and plasma level of VEGFA was determined. The relationship between SNP of VEGFA and plasma level of VEGFA was analyzes. Results: Alleles of 4 loci, rs833068, rs3024994, rs3024997 and rs3025000 showed different distribution between Han and Li nationality. rs3025030 and rs3025033 were significantly correlated with risk of COPD in Han nationality, and could increase the plasma level of VEGFA. Conclusions: SNP of VEGFA has different effect on plasma level of VEGFA between Han and Li nationality.
