摘  要:目的:基于网络药理学,初步探讨预测黄芪抗肝癌的有效成分及潜在作用靶点。方法:通过建立黄芪药物靶点数据集、肝癌疾病靶点数据集,构建黄芪抗肝癌的"成分-靶点-疾病"交互网络图,并通过可视化分析,预测黄芪抗肝癌的有效成分及潜在作用靶点。结果:黄芪可能是通过直接作用于白细胞介素6、细胞肿瘤抗原p53、肝药酶CYP3A4、丝裂原活化蛋白激酶14、细胞周期素A2及肝药酶CYP2A6等蛋白靶点,并通过调节核苷酸结合寡聚化结构域样受体信号通路、肿瘤坏死因子信号通路及鞘脂信号通路等多条通路达到其治疗目的。结论:本研究通过网络药理学初步揭示了黄芪抗肝癌的潜在调控网络,为其后续实验研究及临床应用提供了参考。OBJECTIVE:To probe into the effective components and potential targets of Astragalus in anti-liver cancer based on network pharmacology.METHODS:By establishing a dataset of the drug target of Astragalus and liver cancer disease target dataset,the"ingredients-target-disease"interactive network of Astragalus in treatment of liver cancer was constructed.The active ingredients and potential effects of Astragalus to anti-liver cancer were predicted by visual analysis.RESULTS:Astragalus may act on protein targets such as interleukin-6,cell tumor antigen p53,CYP3A4,mitogen-activated protein kinase 14,cyclin A2,and CYP2A6 directly and simultaneously regulating nucleotide-binding oligomerization domain-like receptor signaling pathways,TNF signaling pathways,sphingolipid signaling pathways and other pathways to achieve their therapeutic goals.CONCLUSIONS:This study preliminarily revealed the potential regulatory network of Astragalus against liver cance through network pharmacology,which provided a reference for the follow-up experimental research and clinical application of Astragalus.
