摘  要:随着人们生活水平的提高,紫心甘薯因其具有较高的营养和保健价值越来越受到人们的欢迎.本研究利用聚丙烯酰胺凝胶电泳筛选到2对多态性好的SSR引物XZSSR15和XZSSR63.利用毛细管电泳技术对筛选到的引物进行多态性检测,共检测出35个位点,每个位点均具有多态性,每对引物平均多态性位点数17.5个,引物平均多态信息含量(PIC)为0.604 3,Shannon多样性指数(I)为0.354 5,Neil多样性指数(H)为0.223 6.利用2对引物构建73份紫心甘薯品种的SSR指纹图谱,通过分析遗传相似性系数和0,1指纹图谱,发现2对SSR引物可以将73份紫心甘薯品种完全区别开来.Purple-fleshed sweetpotato(PFSP)becomes more and more popular due to its high nutritional value for people’s health.In this study,2 polymorphic SSR primers,XZSSR15 and XZSSR63,were selected by polyacrylamide gel electrophoresis.The polymorphism of the selected primers were detected by capillary electrophoresis,and there was a total of 35 loci which were all polymorphic.The average polymorphism of each pair of primers was 17.5,and the average primer polymorphism information content(PIC)was 0.604 3.The Shannon information index(I)averaged 0.354 5,and the Neil diversity index(H)averaged 0.223 6.The SSR fingerprints of 73 PFSP cultivars were constructed by using these 2 primers pairs.The analysis of genetic similarity coefficients and the 0,1 fingerprints indicted that 73 PFSP cultivars could be distinguished completely by XZSSR15 and XZSSR63 SSR primers.
