摘  要:目的观察脑出血前后灯盏花素腹腔注射的大鼠脑损伤程度。方法 96只SD雄性大鼠随机分为A(假手术组)、B(脑出血组)、C(低剂量灯盏花素干预组)、D组(高剂量灯盏花素干预组),每组各24只。B、C、D组采用尾状核定位注射制作脑出血模型,A组仅针刺尾状核定位注射。C、D组分别于造模前1 h及造模1、6、12 h时腹腔注射50、100 mg/kg的灯盏花素,B组分别于造模前1 h及造模1、6、12 h时腹腔注射1 m L生理盐水,A组分别于同时腹腔注射1 m L生理盐水。造模24 h时取各组大鼠,断头取脑,利用大体组织估算各组脑出血体积、干湿重法测定各组脑组织含水量,取血肿周围脑组织,免疫组化法检测各组血肿周围脑组织炎症因子核因子κB(NF-κB)p65,采用Western blotting法检测各组脑组织通路蛋白NF-κB p65、Notch家族受体蛋白1(Notch1)。结果与A组比较,B、C、D组大鼠脑出血体积、脑组织干湿比重增加(P均≤0. 05),与B组比较,C、D组大鼠脑出血体积、脑组织干湿比重减小,且D组减少更明显(P均≤0. 05)。与A组比较,B、C、D组血肿周围脑组织NF-κB p65相对表达量增加(P均≤0. 05),与B组比较,C、D组血肿周围脑组织NF-κB p65的相对表达量降低(P均≤0. 05),与C组比较,D组血肿周围脑组织NF-κB p65的相对表达量降低(P≤0. 05)。与A组比较,B、C、D组脑组织脑组织NF-κB p65、Notch1的相对表达量增加(P均≤0. 05),与B组比较,C、D组脑组织NF-κB p65、Notch1的相对表达量降低(P均≤0. 05),与C组比较,D组脑组织NF-κB p65、Notch1的相对表达量降低(P≤0. 05)。结论灯盏花素腹腔注射可减轻脑出血模型大鼠早期脑损伤程度,其机制可能为抑制NF-κB p65、Notch1的表达。ObjectiveTo observe the effect of breviscapine on early brain injury in rat models with cerebral hemorrhage and to explore its possible mechanism.MethodsNinety-six male SD rats were randomly divided into groups A(sham),B(cerebral hemorrhage group),C(low-dose breviscapine intervention group),and D(high-dose breviscapine intervention group),with 24 rats in each group.Rats in groups B,C and D were injected with caudate nucleus to make intracerebral hemorrhage models.Rats in the group A only underwent acupuncture at the same site without drug injection.Four groups were injected with 1 mL physiological saline with or without breviscapine(group A:saline only;group B:saline only;group C:saline with 50 mg/kg breviscapine;group D:saline with 100 mg/kg breviscapine)at four time points(1 h before model establishment;1,6,and 12 h after the model was established).Brains were dissected at 24 h after cerebral hemorrhage and brain tissue samples were collected.The gross tissue volume of cerebral hemorrhage was estimated by gross tissue,and the water content of brain tissue was measured by wet weight method(WBC).Immunohistochemistry was used to detect the inflammatory factor nuclear factor-κB(NF-κB)p65 in the brain tissue of each group,and Western blotting was used to detect the pathway protein NF-κB p65 and Notch1 in each group.ResultsCompared with the group A,the volume of cerebral hemorrhage and the dry weight of brain tissues in the groups B,C and D increased(all P<0.05).Compared with the group B,the volume of cerebral hemorrhage and the ratio of dry and wet brain tissues in the groups C and D decreased,and the decrease in the group D was more significant(all P≤0.05).Compared with the group A,the relative expression of NF-κB p65 in the brain tissues of the groups B,C and D increased(all P<0.05).Compared with the group B,the relative expression of NF-κB p65 in the brain tissues of the groups C and D decreased(both P<0.05),and compared with the group C,the relative expression of NF-κB p65 in the brain tissues of group D
