摘  要:利用表达L-谷氨酸氧化酶(LGOX)的工程菌进行全细胞催化L-谷氨酸钠合成α-酮戊二酸,考察了各无机离子对酶活的影响,结果表明1mM Mn2+和Mg2+分别对酶活有22%和15%的提高,而Fe2+、Cu2+和Ba2+对酶活有不同程度的抑制。正交实验设计考察催化反应中的影响因素,包括温度、pH、投菌量(固定底物浓度为100g/L)、过氧化氢酶添加量,结果表明最优工艺为温度为30℃、pH值为6.5、投菌量2%、过氧化氢添加量为2%,反应15h,摩尔转化率为99%,该工艺结果对α-酮戊二酸的酶催化工艺有很强的指导作用。The engineering bacteria expressing L-glutamate oxidase(LGOX)were used to synthesize alpha-ketoglutarate from L-glutamate by whole-cell catalysis.The effects of inorganic ions on the activity of the enzyme were investigated.The results showed that1mM Mn2+and Mg2+increased the activity of the enzyme by22%and15%,respectively,while Fe2+,Cu2+and Ba2+inhibited the activity of the enzyme to varying degrees.The orthogonal experimental design was used to investigate the influencing factors in the catalytic reaction,including temperature,pH,bacterial dosage(fixed substrate concentration is100g/L),catalase dosage.The results showed that the optimum process was temperature30℃,pH value6.5,bacterial dosage2%,hydrogen peroxide dosage2%,reaction time15h,molar conversion99%.It has a strong guiding role in the enzymatic catalytic process of alpha ketopglutaric acid.
