摘 要:目的研究不同剂量镇肝熄风汤对自发性高血压大鼠(SHR)腹主动脉血管组织中Ang Ⅱ 1型受体(AT1R)蛋白和Ang Ⅱ 1型受体(AT1R) mRNA表达的影响。方法 60只24周龄的SHR大鼠随机分为:模型组、镇肝熄风汤低剂量组、镇肝熄风汤中剂量组、镇肝熄风汤高剂量组、复方罗布麻组,每组12只; 12只24周龄的同源雄性魏-凯二氏大鼠(WKY)作为:正常对照组。每组灌胃给药45天后,Western Blotting检测方法,检测腹主动脉血管组织中AT1R蛋白表达;实时聚合酶链反应(RT-PCR)检测方法,测定大鼠AT1R mRNA基因表达。结果与模型组比较,正常组、镇肝熄风汤中剂量、高剂量组和复发罗布麻组AT1R蛋白表达和AT1R mRNA表达明显降低(F=863. 465,P <0. 01)。结论镇肝熄风汤可以调低SHR大鼠腹主动脉血管组织中AT1R蛋白表达和AT1R mRNA表达,减少SHR大鼠的血管收缩能力,起到有效降低SHR大鼠血压作用。ObjectiveTo investigate the effect of different doses of zhen-gan-xi-feng decoction on expressions of AT1R mRNA and protein in the abdominal aortic vascular tissue of spontaneously hypertensive rats(SHR).Methods A total of 60 male SHR,24 weeks old,were randomly divided into groups of model,low dose of decoction,medium dose of decoction,high dose of decoction and the compound apocynum,12 in each group.Twelve homologous male rats(WKY)were used to serve as the normal control group.After gavage of the animals for 45 days,western blotting and RT-PCR were used to detect the AT1R protein and mRNA expressions in the abdominal aortic vascular tissue,respectively.Results Compared with the model group,the AT1R protein and mRNA expressions in the other groups were significantly decreased(F=863.465,P<0.01).Conclusion Thezhen-gan-xi-feng decoction can inhibit the expression of AT1R protein and mRNA in the abdominal aortic vascular tissue of SHR and reduce the vasoconstriction,thus,plays a role in lowering blood pressure of the animals.
