摘 要:目的研究姜黄素对人单核细胞(THP-1)源性巨噬泡沫细胞B类1型清道夫受体(SR-B1)表达的影响。方法使用不同浓度姜黄素(12.5、25、50 mg/L)作用于THP-1巨噬泡沫细胞,观察三组不同浓度姜黄素作用的THP-1源性巨噬泡沫细胞SR-B1表达情况。结果仅50 mg/L姜黄素作用24 h细胞存活率低于对照组,差异有统计学意义(P<0.05);其他浓度及时间姜黄素组细胞存活率与对照组比较,差异无统计学意义(P>0.05)。50 mg/L姜黄素组SR-B1、TRAF6、NF-κB以及IRAK1 mRNA表达水平明显低于其他组,差异有统计学意义(P<0.05);25 mg/L姜黄素组上述指标表达水平低于12.5 mg/L姜黄素组和对照组,差异有统计学意义(P<0.05);12.5 mg/L姜黄素组与对照组上述指标表达水平比较,差异无统计学意义(P>0.05)。结论一定浓度姜黄素可抑制THP-1源性巨噬泡沫细胞中SR-B1、TRAF6、NF-κB以及IRAK1 mRNA的表达,从而减少泡沫细胞,达到抗动脉粥样硬化的目的。Objective To investigate the effect of curcumin on the expression of class B type 1 scavenger receptor(SR-B1)in human monocyte-derived(THP-1)-derived macrophage foam cells. Methods Different concentrations of curcumin(12.5, 25, 50 mg/L) were used to treat THP-1 macrophage foam cells, and the expression of SR-B1 in THP-1 derived macrophage foam cells treated with different concentrations of curcumin was observed. Results The survival rate of only 50 mg/L curcumin for 24 h was lower than that of the control group, and the difference was statistically significant(P<0.05). There was no significant difference between the other concentrations and time of curcumin group and the control group(P>0.05). The expression levels of SR-B1, TRAF6, NF-κB and IRAK1 mRNA in the 50 mg/L curcumin group were significantly lower than those in the other groups, and the difference was statistically significant(P<0.05). The expression level of the above indicators in the 25 mg/L curcumin group was lower than that of the 12.5 mg/L curcumin group and control group, and the difference was statistically significant(P<0.05). There was no statistically significant difference between the 12.5 mg/L curcumin group and the control group in the above indicators (P>0.05). Conclusion Certain concentration of curcumin can inhibit the expression of SR-B1, TRAF6, NF-κB and IRAK1 mRNA in THP-1 derived macrophage foam cells, thereby reducing foam cells and achieving anti-atherosclerosis.
