摘 要:目的:研究组分配伍四逆汤对被动型Heymann肾炎大鼠的治疗作用。方法:兔皮内多次免疫制备兔抗鼠Fx1A抗血清,然后大鼠尾静脉注射Fx1A抗血清,建立被动型Heymann肾炎(PHN)模型。将实验大鼠随机分为4组,为正常组、模型组、肾衰宁组和组分配伍四逆汤组。造模成功后,每天用相应的药物进行灌胃给药治疗,持续8周。第8周末收集各组大鼠尿液用于检测大鼠24 h尿蛋白。给药治疗结束后,收集各组大鼠的血清用于检测血清中的肌酐(Scr)、尿素氮(BUN)、白蛋白(Alb)、总胆固醇(TC)和甘油三酯(TG)等指标;取肾脏,进行HE染色,观察组分配伍四逆汤对PHN大鼠病理损伤的影响。结果:连续灌胃8周后,组分配伍四逆汤可以明显降低PHN大鼠24 h尿蛋白含量(P<0.01),显著降低PHN大鼠血清Scr(P<0.01)、BUN(P<0.05)、TC(P<0.01)以及TG(P<0.01)水平,并显著升高PHN模型大鼠血清Alb(P<0.01);HE染色结果表明,组分配伍四逆汤能减轻PHN模型大鼠肾组织的病理变化。结论:组分配伍四逆汤对被动型Heymann肾炎大鼠具有改善作用。Objective:To investigate the therapeutic effect of component compatibility Sini Decoction on passive heymann nephritis in rats.Methods:Rabbits were given intradermal immunization for many times to obtain anti-Fx1 A serum,and then PHN model in rats was established by tail intravenous injection of anti-Fx1 A serum.The experimental rats were randomly divided into four groups:the normal group,model group,shenshuaining group,component compatibility Sini Decoction group.After the success of modeling,each rat received intragastric administration for eight weeks.At the end of the eighth week,the total of urine was collected for the detection of 24-hour urinary protein.At the end of treatment,the serum was collected for the detection of serum creatine(Scr),blood urea nitrogen(BUN),serum Albumin(Alb),total cholesterol(TC)and triglyceride(TG).The pathological changes of the renal tissue were observed using hematoxylin-eosin to observe the effect of component compatibility Sini Decoction on PHN rats.Results:After the intragastric administration for eight weeks,component compatibility Sini Decoction could obviously reduce the level of 24-hour urinary protein in PHN rats(P<0.01),significantly reduce the levels of Scr(P<0.01),BUN(P<0.01),TC(P<0.01)and TG(P<0.01)in PHN rats,and increase the level of Alb(P<0.01).HE results indicated that component compatibility Sini Decoction could reduce the pathological changes of renal tissue in PHN rats.Conclusion:Component compatibility Sini Decoction has notable improvement for PHN in rats.
