柔毛淫羊藿中淫羊藿苷含量测定及最佳采收期研究

摘　　要：目的建立测定不同采收期柔毛淫羊藿中淫羊藿苷含量的高效液相色谱(HPLC)法,为合理采收和开发利用提供参考。方法色谱柱为Agilent C_(18)柱(250 mm×4. 6 mm,5μm),流动相为乙腈-0. 1%磷酸溶液(25∶75,V/V),流速为1. 0 mL/min,柱温为30℃,检测波长为270 nm。结果淫羊藿苷进样量在58. 40～1 168. 00 ng范围内与峰面积线性关系良好,r=0. 999 9(n=5);平均加样回收率为95. 59%,RSD为1. 77%(n=6);不同采收期的柔毛淫羊藿中淫羊藿苷含量差异较大,其中7月最高(0. 723 9%),2月最低(0. 223 5%)。结论该方法操作简单、快速、准确,可为确定淫羊藿药材的最佳采收时间提供参考,以合理开发利用淫羊藿药材。Objective To establish an HPLC method for the content determination of icariine in Epimedium pubescens in different harvest seasons,in order to provide reference for rational harvesting and exploitation.Methods The chromatographic column was Agilent Cis column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-0.1% phosphoric acid(25-75,V/V),the flow rate was 1.0 mL/min,the column temperature was 30℃,and the detection wavelength was 270 nm.Results The icariine showed good relationship in the range of 58.40-1168.00 ng,r=0.9999(n=5).The average recovery rate was 95.59%,RSD was 1.77%(n=6).The content of icariine in Epimedium pubescens collected in different harvest seasons was quite different,with the highest in July(0.7239%)and the lowest in February(0.2235%).Conclusion The method is simple,rapid and accurate.It can provide reference for making sure the optimal harvest season of Epimedium pubescens,so as to ensure the rational development and utilization of Epimedium pubescens.
