摘 要:目的探究右美托咪定与育亨宾对慢性应激小鼠抑郁的影响:方法将筛选出的40只昆明小鼠随机分为4组(n=10):对照组、模型组、右美托咪症组、育亨宾组。除对照组外,每组均通过慢性应激建立抑郁模型.采用旷场实验和强迫游泳实验进行行为学研究,并用ELISA测定小鼠脑内去甲肾上腺素(NE)和α2A受体的含量结果与对照组相比,模型组的旷场总运动距离和旷场运动速度均显著减少,强迫游泳不动时间明显增加,脑内蓝斑核的NE含量明他减少,而α2A受体的含量显著增高,差异有统计学意义(P<0.05)。与模型组相比,右美托咪定组的旷场总运动距离和旷场运动速度均显著减少,强迫游泳不动时间明显增加,脑内蓝斑核的NE含量降低,α2A受体含量显著增加,差异有统计学意义(P<0.05);与模型组相比,育亨宾组的旷场总运动距离和旷场运动速度均显著增加,强迫游泳不动时间明显减少,脑内蓝斑核的NE含量显著增加,α2A受体含量显著降低,差异有统计学意义(P<0.05)结论右美托咪定具有一定加重抑郁的作用,而育亨宾具有一定减轻抑郁的作用。Objective To observe the effects of dexmedetomidine and yohimbineon on mice with chronic stress-induced depression. Methods A total of 40 Kunming mice were raiulornly divided into four groups (n=10 ): a control group, a model group, a ilexmedetoniidine group and a yohinihine group. Chronic unpredictable stimuli were adopted to establish a model of depression in mice. Then, the open field test and forced swimming test were used to research the behavior of these mice. The content of α2A receptors and norepinephrine (NE) in the locus coeruleus (LC ) of mice were measured by ELISA. Results Compared with the control group, the model group presented remarkable decreases in the total distance and speed of the open field movement and marked increases in the duration of immobility in the forced swimming test, as well as significantly reduced NE content and increased α2A receptor levels in the LC (P<0.05 ). Compared with the model group, the dexmedetomidine group presented remarkable decreases in the total distance ancl speed of the open field niovrment and marked increases in the duration of immobility in the forced swimming test, as well as significantly reduced NE content and increased α2A receptor levels in the LC (P<0.05 ). Compared with the model group, the yohimbine group presented remarkable increases in the lolal distance and speed of the open field movement and marked decreases in the duration of immobility in the forced swimming test, as well as significantly increased NE content and reduced α2A receptor levels in the LC (P<0.05 ). Conclusions Dexmedetomidine can aggravate depression, and yohimbine can alleviate depression.
