摘  要:目的桃红四物汤是养血活血的经典复方,本文运用网络药理学方法探究其"成分-靶标-疾病"的关系。方法运用中药系统药理学数据库(TCMSP)收集桃红四物汤活性成分,多个数据库联用检索查询成分和疾病对应的靶标,并利用Cytoscape 3.5.1软件对成分靶标与疾病靶标进行网络拓扑学分析,构建"成分-靶标-疾病"的可视化网络拓扑关系图。通过基因功能富集平台GATHER在线分析信号通路。结果共检索到桃仁四物汤全方730个化学成分,根据类药性筛选出潜在活性成分61个,从HPO数据库中获得可作用于冠心病的潜在靶标90个。分析可视化网络拓扑图,共有节点39个、328条边。得到4个共同靶标APOE、APOB、CYP7A1、ABCA12和5个关键通路Alzheimer's disease、MAPK signaling pathway、Adherens junction、Huntington's disease、Jak-STAT signaling pathway。结论本研究筛选了桃红四物汤治疗冠心病的关键靶标和通路,为桃红四物汤用于临床治疗提供了科学依据。Objective To identify the active constituents and targets of Taohong Siwu decoction a classical prescription for nourishing blood and promoting blood circulation, and to determine the relationship between"compound-target-disease" with network pharmacology. Methods Traditional Chinese medicine system pharmacology(TCMSP) database and the related literatures were applied to screen out the potential active compounds in Taohong Siwu decoction. Multiple databases were used to search the targets of the potential active compounds and coronary heart diseases. The compounds related targets and coronary heart diseases related targets were visualized by Cytoscape 3.5.1, and a visual network topology map of the "compound-target-disease" was constructed. The functional targets and mechanism pathways were determined with the gene function enrichment platform GATHER. Results Totally 61 active compounds in Taohong Siwu decoction were screened out from 730 compounds and 90 coronary heart disease related targets were obtained. From the visual network topology map, 4 targets(APOE, APOB, CYP7 A1, and ABCA12) were obtained. The GATHER database suggested 5 key mechanism pathways(Alzheimer’s disease, MAPK signaling pathway, Adherens junction, Huntington’s disease, and Jak-STAT signaling pathway). Conclusion The key targets and signaling pathways of Taohong Siwu decoction have been screened in the treatment of coronary heart diseases, providing a theoretical basis for the further study of Taohong Siwu decoction.
