摘  要:目的:观察电针双侧"足三里"穴对脾气虚模型大鼠骨骼肌组织PGC-1α/SIRT3信号通路表达的影响。方法:将32只SD雄性大鼠随机分为空白组、脾气虚组、足三里组、非经非穴组。除空白组外,其余3组均采用劳倦过度和不规则饮食复合法建立脾气虚证大鼠模型。造模成功后,足三里组给予电针双侧"足三里"穴治疗,非经非穴组予以电针双侧非经非穴点治疗,每日1次,每次20 min,共治疗7 d。观察各组大鼠的一般状态,采用大小鼠抓力测定仪测量大鼠前肢抓力;采用荧光定量PCR法检测大鼠骨骼肌PGC-1α、ERRα、SIRT3及SOD2的mRNA表达水平;采用Western blot检测大鼠骨骼肌PGC-1α、ERRα、SIRT3及SOD2的蛋白表达水平。结果:与空白组相比,脾气虚组和非经非穴组大鼠形体消瘦,体质量下降,进食量减少,皮毛枯槁脱落,懒动,大便稀溏;抓力显著下降(P <0. 05);骨骼肌组织中PGC-1α、ERRα、SIRT3及SOD2的mRNA和蛋白表达水平均明显降低(P <0. 05)。与脾气虚组相比,足三里组大鼠活动性增加,进食量增加,体质量略增加,被毛掉落减少,便溏情况有所好转;大鼠抓力明显增大(P <0. 05);骨骼肌组织中PGC-1α、ERRα、SIRT3及SOD2的mRNA和蛋白表达水平均明显升高(P <0. 05),非经非穴组未见显著性差异(P> 0. 05)。结论:电针"足三里"穴可以调节脾气虚大鼠骨骼肌内PGC-1α/SIRT3信号通路的异常表达,参与线粒体生物合成及抗氧化的调控作用进而发挥健脾益气作用。Objective: To observe the effect of electroacupuncture on bilateral "Zusanli"( ST36) point on the expression of PGC-1 alpha/SIRT3 signaling pathway in skeletal muscle tissue of rats with spleen Qi deficiency. Methods: Thirty-two male SD rats were randomly devided into 4 groups: normal group,spleen Qi deficiency group,acupuncture group and sham acupuncture group( n = 8). The model of spleen Qi deficiency syndrome was established by the complex factors of improper diet and overstrain. Electroacupuncture( 2 Hz/15 Hz,0. 5 mA) was applied to bilateral"Zusanli"( ST36) in acupuncture group and sham acupoint in sham acupuncture group for 20 min,once a day for 7 days. The general condition of rats in each group was observed. The levels of mRNA expressions of PGC-1 alpha,ERRα,SIRT3 and SOD2 in rat skeletal muscle tissue were detected by fluorescence quantitative PCR. Western blot was used to detect the protein expression levels of ERRα,SIRT3 and SOD2 in rats’ skeletal muscle tissue. Results: Compared with the control group,rats in spleen Qi deficiency and sham acupuncture group were weight loss,food intake reduced,haggard fur off,lazy and loose stool;grasping force decreased significantly( P < 0. 05). The mRNA and protein expression of PGC-1 alpha,ERRα,SIRT3 and SOD2 in skeletal muscle decreased significantly( P < 0. 05). Compared with spleen Qi deficiency group,the acupuncture group’ s food intake increased,body weight increased slightly,hair dropping,loose stools and the situation improved. The rat grasping force increased significantly( P < 0. 05). The mRNA and protein expressions of PGC-1 alpha,ERRα,SIRT3 and SOD2 in skeletal muscle increased obviously( P < 0. 05). The general state and grasping force of rats in sham acupuncture group were not significantly improved( P > 0. 05),and the gene and protein expression was not significantly changed( P > 0. 05). Conclusion: Electroacupuncture at "Zusanli"( ST36) can modulate the abnormal expressions of PGC-1 alpha/SIRT3 signaling pathway in skeletal muscle of spleen Q
