摘 要:目的:探讨滋阴养血安神方对氯苯丙氨酸(PCPA)诱导的睡眠剥夺小鼠脑内多巴胺(DA)含量的影响。方法:利用腹腔注射PCPA制造睡眠剥夺模型,分设滋阴养血安神方18.25、36.5、73、146、292、584 g/kg剂量给药组,与模型组相比较,以阈上剂量戊巴比妥钠诱导的睡眠时间摸索滋阴养血安神方给药剂量。腹腔注射PCPA制造睡眠剥夺模型,分6组,空白对照组、模型组、滋阴养血安神方低剂量组、滋阴养血安神方中剂量组、滋阴养血安神方高剂量组、地西泮组(1.5 mg/kg),造模并给药7 d后,采用ELISA方法检测下丘脑与海马DA含量及Western-blot方法检测小鼠下丘脑DA蛋白含量。结果:与模型组对比,滋阴养血安神方用药组小鼠下丘脑DA含量明显低于模型组,与模型组对比有显著性差异(P<0.01)。滋阴养血安神方用药组海马DA含量高于模型组,尤其是高剂量组,与模型组对比有显著性差异(P<0.05)。Western-blot方法检测下丘脑D1受体含量,与模型组对比,滋阴养血安神方各给药组D1受体降低,尤其是高剂量组,与模型组对比有显著性差异(P<0.05)。结论:滋阴养血安神方具有镇静、催眠作用,对失眠症有一定的治疗作用,其对小鼠睡眠的影响机制之一可能是通过降低下丘脑DA含量及D1受体实现的。滋阴养血安神方对海马区DA含量有影响,可能会影响小鼠认知行为。Objective:By detecting the contents of dopamine(DA)in mice brains to observe Ziyin Yangxue Anshen Decoction on sleep deprivation mice induced by para-chlorophenylalanine(PCPA).Methods:The model was established by receiving PCPA,the dose of Ziyin Yangxue Anshen Decoction was 18.25,36.5,73,146,292 and 584 g/kg respectively,and compared with the model group,the drug dose of Ziyin Yangxue Anshen Decoction was explored with the sleep time induced by the upper threshold dose of pentobarbital sodium. The model was established by receiving PCPA,and six groups such as blank control group,model group,the low dose group of Ziyin Yangxue Anshen Decoction,the mediumdose group of Ziyin Yangxue Anshen Decoction,the high dose group of Ziyin Yangxue Anshen Decoction,diazepam group(1.5 mg/kg)were established. After each drug administration group was given 7 days,DA content in the hypothalamus and hippocampus were detected by ELISA and D1 receptors content was detected by Western-blot. Results:Compared with the model group,the content of DA in the hypothalamus of mice treated with Ziyin Yangxue Anshen Decoction groups were significantly lower than that of the model group(P<0.01). Compared with the model group,the contents of hippocampal DA in the Ziyin Yangxue Anshen Decoction groups were higher than those in the model group,especially in the high-dose group(P<0.05). Compared with model group,contents of the hypothalamus D1 receptors treated by Ziyin Yangxue Anshen Decoction lower than that of the model group detecting by Western blot method,especially in high dose group,there are significant differences between them(P<0.05). Conclusion:Ziyin Yangxue Anshen Decoction have sedative and hypnotic effect,has a certain therapeutic effect on insomnia,one of its impact on sleep in mice mechanism may be through the implementation of the lower brain content of DA and D1 receptors. Ziyin Yangxue Anshen Decoction have an effect on the content of DA in the hippocampal area,which might affect the cognitive behavior of mice.
