背俞穴埋线法改善围绝经期大鼠脂质代谢异常机制研究  

Effect of Back-Shu Acupoint Catgut-embedding Therapy on Lipid Metabolism and Its Possible Mechanism in the Perimenopausal Rats

摘　　要：目的:探讨穴位埋线法通过调节下丘脑-垂体-性腺轴改善围绝经期大鼠脂质代谢异常。方法:利用双侧卵巢切除(OVX)的方法制备围绝经期大鼠模型,SD大鼠随机分为假手术组、模型组和埋线组,每组10只。比较各组大鼠体质量、Lee's指数、腹腔脂肪重量、血清中雌二醇(E2)、促卵巢刺激激素(FSH)、促黄体生成素(LH)、下丘脑促性腺激素释放激素(GnRH)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL)和高密度脂蛋白(HDL)、瘦素及脂联素表达变化。结果:与假手术组相比,模型组大鼠E2水平明显下降,FSH、LH和GnRH水平明显升高;血清中TC、TG、LDL含量均升高,而HDL含量下降;瘦素含量升高而脂联素含量降低,具有显著性差异(P<0.05,P<0.01)。穴位埋线能够降低大鼠体质量、Lee's指数、腹腔脂肪重量;增加大鼠血清中E2水平,降低FSH、LH和GnRH表达水平;降低血清中TG、LDL含量,升高HDL含量;降低瘦素含量,升高脂联素含量,具有显著性差异(P<0.05,P<0.01)。结论:穴位埋线法能够通过调节下丘脑-垂体-性腺轴改善围绝经期大鼠雌激素紊乱、能量代谢和脂质代谢异常。Objective:To investigate the effect of acupoint catgut-embedding therapy on lipid metabolism in perimenopausal rats by regulating hypothalamus pituitary sexual gland. Methods:SD rats were randomly divide into the sham group,the model group and the acupoint catgut-embedding therapy group,10 mice in each group. In this study,the perimenopausal model in rats was established by ovariectomy. Compared the differences in rats weight,Lee’s index,abdominal cavity fat weight;serum estradiol(E2),promote ovarian stimulating hormone(FSH),luteinizing hormone(LH),hgonadotropin releases hormones(GnRH),total cholesterol(TC),triglyceride(TG),low density lipoprotein(LDL)and high-density lipoprotein(HDL)levels;expressions of leptin and adiponectin by ELISA. Results:Compared with the sham group,the level of E2 in the model group was significantly decreased,and FSH,LH and GnRH levels in the model group were significantly increased;and TG,LDL levels in the model group were increased,and HDL levels in the model group were decreased;the expression of leptin in the model group was increased,in contrast,adiponectin expression in the model groupwas lower,there were significant difference between sham and model group(P<0.05,P<0.01). Catgut-embedding at acupoints can reduced the weight of rats,Lee’s index and abdominal fat weight;increased the level of E2 in serum,decreased the expression levels of FSH,LH and GnRH;decreased the content of TG,LDL and HDL in serum;decreased the content of leptin and increased the content of adiponectin,with significant difference(P<0.05,P<0.01). Conclusion:The acupoint catgut-embedding therapy improve the disorder of estrogen,energy metabolism and lipid metabolism in perimenopausal rats by regulating hypothalamus pituitary sexual gland.
