摘  要:为了研究四环素标记鱼类的效果及其对抗氧化能力的影响,设置100、150、250 mg/L浓度梯度的盐酸四环素(TCH)溶液和不同浸泡时间(18 h和24 h)对毒理实验常用对象斑马鱼(Danio rerio)进行浸泡标记,通过观察TCH浸泡标记后30 d内斑马鱼耳石的标记效果以及浸泡后第1、3、9、15 d时鱼体内三种抗氧化酶(SOD、CAT、GSH-Px)的活性变化,综合分析其标记鱼类的效果和安全性。结果显示,在150 mg/L和250 mg/L浓度的TCH溶液中浸泡18 h或24 h,斑马鱼耳石标记率可达100%,且耳石上可见清晰的黄色标记轮纹。浸泡18 h时,中高浓度组(150 mg/L和250 mg/L)斑马鱼的抗氧化酶活性在浸泡后的第3 d显著增加。浸泡24 h时,各浓度组的抗氧化酶活性在第1 d显著升高,250 mg/L浓度组的SOD和GSH-Px活性分别在第1 d和第9 d显著低于对照组,各组的抗氧化酶活性均在第15 d时恢复为对照组水平。结果表明,100 mg/L和150 mg/L浓度的TCH浸泡液对斑马鱼的抗氧化酶活性起诱导作用,250 mg/L浓度的TCH溶液浸泡,对其抗氧化酶活性产生了短期的抑制作用并可能对机体产生可恢复的氧化损伤。Danio rerio were commonly used for immersion marking in toxicological experiments to study the effects of marking fish and its anti-oxidase activity. This study set different concentration gradient of tetracycline hydrochloride ( TCH) solution of 0 mg/L, 100 mg/L, 150 mg/L, 250 mg/ L and different soaking times ( 18 h and 24 h). By observing The marking effects of D rerio otolith on the 1 st , 3 rd , 6 th , 9 th , 15 th and 30 th day after TCH immersion marking were observed in this study. Meanwhile, the activity changes of three antioxidant enzymes ( SOD, CAT, GSH-Px) in fish were detected. Comprehensive analysis of the efficacy and safety of its labeled fish. The results showed that the D rerio otolith marking rates were 100% when immersed in 150 mg/L and 250 mg/L TCH solution for 18 h or 24 h. A clear yellow marking wheel pattern was observed on the otolith. The antioxidant enzyme activity of D rerio in the medium-high concentration treatment groups ( 150 mg/L, 250 mg/L) were significantly increased on the 3 rd day after soaking at 18 h. When the soaking time was 24 h, the activities of antioxidant enzymes in each concentration group increased significantly on the 1 st day. The activities of SOD and GSH-Px in the 250 mg/L group were significantly lower than those in the control group on the 1 st and 9 th day, respectively. The antioxidant enzyme activities of each group returned to the control level at the 15th day. The results showed that TCH soaking solutions at concentrations of 100 mg/L and 150 mg/L induced the antioxidant enzyme activity of Drerio . Soaking with TCH solution at concentration of 250 mg/L produced a short-term inhibitory effect on antioxidant enzyme activity and may cause recoverable oxidative damage to the body.
