摘 要:本研究旨在分析颈静脉短期滴注化学合成心房利钠肽(ANP)和树眼镜蛇属利钠肽(DNP)嵌合肽AD-NP对绵羊血脂生化指标的影响。试验选取8只体重相近、健康状况良好的1.5岁左右阿勒泰大尾羊母羊为试验动物,采用自身对照试验设计,对照期静脉滴注生理盐水100 mL,试验期根据绵羊体重滴注1.125μg/kg AD-NP的生理盐水100 mL,滴注持续时间45 min。在第4天静脉滴注开始前、开始后15、30、45、60、90和120 min以及滴注结束第2天和第5天12:00采集空腹静脉血液样本以测定血脂生化指标。结果显示,滴注AD-NP对阿勒泰大尾母羊血液中低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C)均无显著影响(P>0.05)。在滴注AD-NP 45 min时血液中甘油三脂(TG)含量较对照期降低26.58%(P<0.05),在滴注30 min时非酯化脂肪酸(NEFA)水平升高47.62%(P<0.05)。在滴注开始和45、120 min时,试验期血液中甘油含量较对照期提高25.04%、36.67%和25.86%(P<0.05)。由此可见,以1.125μg/kg的剂量静脉滴注AD-NP 3 d后在促进绵羊释放TG、甘油和NEFA方面具有一定作用。This experiment was conducted to study the effects of short-term intravenous infusion of a novel chimeric natriuretic peptide AD-NP on the physiological and biochemical parameters of Altay sheep. Eight big tail Altay ewes,aged 1.5 years old with similar body weight and good health were selected as test animals. Self-controlled design was adopted. During the control period, all animals received a short-term intravenous infusion of 100 mL saline. Based on the body weigh of sheep, 1.125 μg/kg AD-NP in 100 mL saline was given in each day during the experimental period. Each period lasted for 4 days and the infusion lasted 45 minutes in each day. From the fourth day of each period, the fasting venous blood samples were collected before and after initial infusion of 15, 30, 45, 60, 90 and 120 min, and at 12:00 of 2th and 5th day after infusion terminated to determine lipids concentration.The results showed that concentrations of low-density lipoprotein cholesterol (LDL-C) and high-density lipoprotein cholesterol (HDL-C) were not affected by AD-NP (P >0.05). At 45 min after AD-NP infusion, the content of TG in plasma was decreased by 26.58%(P <0.05), and the level of non-esterified fatty acid (NEFA) was increased by 47.62% at 30 min (P <0.05). Furthermore, the glycerol contents in plasma at the beginning of AD-NP infution and 45,120 minutes after infusion were significantly increased by 25.04%,36.67% and 25.86%, respectively (P <0.05). Therefore, intravenous infusion of AD-NP at the dose of 1.125 μg/kg for three days can promote the release of TG,glycerol and NEFA in sheep.
