摘 要:为分析超表达生长素(IAA)合成基因YUCCA1对草莓果实成熟进程的影响。以‘甜查理’草莓(Fragaria×ananassa Duch.)果实为试验材料,通过外源喷施IAA,分析IAA对草莓果实发育成熟的影响,利用qPCR测定了果实发育过程中YUCCA1表达量,克隆了草莓YUCCA1,获得了其超表达的草莓果实,测定了YUCCA1超表达后果实的生理及分子指标。随着草莓果实成熟IAA含量下降,外源喷施IAA能延迟果实的成熟,并影响了果实着色、软化和香气代谢等生理和分子指标,而IAA运输抑制剂三碘苯甲酸(TIBA)则能促进果实成熟。草莓IAA合成基因YUCCA1在果实发育前期表达量高,后期表达量急剧下降,在成熟期几乎检测不到。在草莓果实中超表达YUCCA1延迟了果实的成熟进程,增加了果实内源IAA、叶绿素含量和果实硬度,但是降低了花色素和脱落酸(ABA)含量,而IAA运输抑制剂TIBA能解除超表达YUCCA1对果实发育的抑制作用。IAA参与了草莓果实的发育成熟调控,负调控了果实成熟进程,可延缓果实衰老。In this study,‘Sweet Charlie’ fruits (Fragaria × ananassa Duch.) were used as test material, the effects of IAA on the development and ripening process of strawberry fruit was analyzed by exogenous spraying IAA. The expression of YUCCA1 was determined by qPCR. The strawberry YUCCA1 was cloned and its over-expressed strawberry fruit was obtained.The physiological and molecular indicators of YUCCA1 over-expressed fruit was determined. The content of IAA decreased with the ripening of strawberry fruit.Exogenous IAA delayed the fruit ripening process,and influenced the fruit physiological and molecular indicators such as fruit anthocyanin,softening and aroma metabolism,whereas IAA transporter inhibitor 2,3,5-Triiodobenzoic acid (TIBA) could promote fruit ripening process. IAA biosynthesis gene YUCCA1 had the highest expression level in earlier fruit development,and then decreased sharply,ultimately its expression was hardly detected in ripening fruit stage.Overexpression of YUCCA1 gene in fruit delayed the process of fruit ripening process,increased the endogenous IAA content,as well as chlorophyll content and fruit hardness,but reduced anthocyanin and ABA content. However,IAA transporter inhibitor TIBA relieved the inhibitory effect of IAA on fruit ripening. IAA was involved in the regulation of fruit development and ripening process,and negatively regulated the ripening process that delayed fruit senescence.TIBA,a IAA transport inhibitor,could relieve the inhibition of over-expression of YUCCA1 on fruit development.
