摘 要:目的检测N-甲基-D-天冬氨酸(N-mehyl-D-aspartate,NMDA)受体亚基NR2B (NMDA-NR2B)阻断剂艾芬地尔对单眼形觉剥夺小鼠初级视皮层内兴奋性递质受体亚基关键期相对表达量的影响,为弱视机制研究提供实验基础。方法选取出生后3~4周健康C57BL/6J小鼠15只,随机分为正常对照组、单眼形觉剥夺组、联合组,每组5只。单眼形觉剥夺组和联合组小鼠右眼褥式缝合6 d,建立单眼形觉剥夺模型,联合组于第1、3、5天腹腔注射艾芬地尔,给药量为10 mg·kg^(-1)。采用qRT-PCR法检测各组小鼠视皮层NR2A mRNA和NR2B mRNA的相对表达量;采用Western blot法检测各组小鼠视皮层NR2A蛋白和NR2B蛋白的相对表达情况。结果与正常对照组相比,单眼形觉剥夺组小鼠左侧视皮层NR2A mRNA和蛋白表达下降,差异均有统计学意义(均为P<0.05),NR2B mRNA和蛋白表达升高,差异均有统计学意义(均为P<0.05);右侧视皮层NR2A mRNA和蛋白表达升高,差异均有统计学意义(均为P<0.05),NR2B mRNA和蛋白表达下降,差异均有统计学意义(均为P<0.05)。联合组、单眼形觉剥夺组小鼠左侧视皮层NR2A mRNA和蛋白相对表达量分别为0.99±0.22、0.75±0.08和0.58±0.44、0.50±0.36,差异均无统计学意义(均为P>0.05);联合组小鼠左侧视皮层NR2B蛋白相对表达量为1.79±0.84,低于单眼形觉剥夺组小鼠(4.83±2.06),差异有统计学意义(P<0.05)。结论 NR2B受体阻断剂艾芬地尔能显著降低NR2B亚基的表达,并阻断单眼形觉剥夺对NR2B基因及蛋白表达的影响,但对NR2A基因及蛋白表达无影响。Objective To investigate the effects of ifenprodil on the expression of N-methy-D-aspartate (NMDA) receptor subunit NR2B (NMDA- NR2B) following monocular form deprivation in visual cortex of mice during the critical period of visual development for providing theoretical evidences for amblyopia. Methods Totally 15 healthy aged 3-5 weeks C57BL/6J mice were randomly divided into normal control group,monocular form deprivation group and the combined group,5 cases in each group.The right eyes of these mice in the monocular form deprivation group and the combined group was sutured for 6 days to establish the model of monocular form deprivation,while the mice in the combined with intraperitoneal injection group were given unilateral intraperitoneal injection of 10 mg·kg^-1 ifenprodil on day 1,3 and 5.qRT-PCR,Western blot were used to detect the mRNA and protein expression of NMDA receptor subunits NR2A and NR2B. Results The expression levels of NR2A mRNA and protein in the left visual cortex of the monocular form deprivation group were lower,but the levels of NR2B mRNA and protein expression were higher as compared with the normal control group(all P<0.05).The levels of NR2A mRNA and protein in the right visual cortex of the monocular form deprivation group,but the levels of NR2B mRNA and protein were lower than those of the normal control group(all P<0.05).The NR2A mRNA,protein in the left visual cortex of the combined group and monocular form deprivation group was 0.99±0.22,0.75±0.08 and 0.58±0.44,0.50±0.36,and the differences were not statistically significant(all P >0.05).The NR2B protein in the left visual cortex of the combined group was 1.79±0.84,which was lower than of the monocular deprivation group (4.83±2.06),and the difference was statistically significant(P<0.05). Conclusion NR2B subunit inhibitor Ifenprodil could suppress the expression of NR2B protein,and eliminate the effect of monocular deprivation on the expression of NR2B subunit,but it had no effect on the expression of NR2A subunit.
