摘  要:目的探索一种稳定的天灸发泡大鼠模型的制作方法。方法将8只雄性SD大鼠麻醉、脱毛,称取0.018 g棉球填充于PU防水空白膏药贴中央。吸取200μL 5%斑蝥酊均匀浸湿棉球后贴敷刺激,2h后加200μL斑蝥酊1次,密封覆盖,4 h后统一收集泡液。通过观察发泡率、泡体大小、泡液量、泡液吸收情况、发泡区域结痂情况、大鼠死亡率情况以及发泡液内总细胞数量,对本方法进行评价。结果 200μL 5%的斑蝥酊发泡率高,泡液量和泡体大小较理想,大鼠死亡率低。结论用200μL 5%的斑蝥酊可以在大鼠皮肤进行稳定发泡,且操作便捷,大鼠死亡率低。Objective To explore a stable method to establish a rat model of formation of blisters in vesiculation moxibustion. Method Eight male Sprague-Dawley (SD) rats were anesthetized and depilated. 0.018 g cotton balls were filled in the center of the PU waterproof blank paste plaster. 5% cantharides tincture 200 μL was drawn to soak the cotton ball evenly and then was applied. Two hours later, 200 μL cantharides tincture was added once and then it was sealed. The blister fluid was collected 4 h later. This method was evaluated by observing the formation rate of blisters, the size of blister, the volume of blister fluid, the absorption of blister fluid, the scab in the area of blisters, the mortality rate of rats and the total number of cells in blister fluid. Result The formation rate of blisters with application of 5% cantharides tincture 200 μL was high, the volume of blister fluid and the size of blister were more ideal, and the mortality rate of rats was low. Conclusion The stable formation of blisters on the skin of rats can be achieved with application of 5% cantharides tincture 200 μL. The operation is convenient and the mortality rate of rats is low.
