摘 要:目的利用体外构建的组织工程皮肤模型进行化学品刺激性检测。方法利用组织工程皮肤构建方法,以牛Ⅰ型胶原为支架,以人的正常皮肤角质形成细胞和成纤维细胞为细胞来源,构建组织工程皮肤模型。在欧洲替代实验确认中心规定的用于皮肤刺激性检测的物质中选取了20种化学品为检测物质。20种化学品取25μl(固体25mg研成粉末,同时加蒸憎水25μl),局部接触检测模型表面15min,利用MT1、方法测定细胞活力,阴性对照加25μl蒸僧水,阳性对照加25μl的5%月桂硫酸钠。结果以相对细胞活力值为判定刺激性的依据,在20种化学品中,构建的检测模型能够分辨16种化学品的刺激性。结论构建的组织工程皮肤模型可以判定化学品的刺激性,可以作为化学品刺激性的检测方法。Objective To undergo chemical irritation detection using a tissue engineering skin model established in vitro.Methods The tissue engineering skin model was constructed using bovine type Ⅰ collagen as scaffold and human normal skin keratinocytes and fibroblasts as cell sources.Twenty chemicals were selected as test substances from the substances specified by the European centre for validation of alternative experiments for skin irritation.During the detection, 25 of 20 chemicals (25 mg solid were ground into powder before added into 25μl distilled water) were contacted with the surface of the test model for 15 min.The cell viability was measured by MTF.Meanwhile, 25μl distilled water was added to the negative control and 25μl of 5 % sodium laurate was added to the positive control.Results Based on the relative cell viability values, 16 chemicals were identified by the reconstructed detection model.Conclusions The tissue engineering skin model determines the irritation of chemicals and can be used as a detection method for chemical irritation.
