针刺对雷公藤所致卵巢功能减退模型小鼠卵巢的影响

摘　　要：目的观察针刺对卵巢功能减退模型小鼠卵巢的保护作用。方法 40只动情周期正常的小鼠随机分为4组,每组10只,分别为空白组、模型组、雌二醇组、针刺组。除空白组每日灌胃等体积生理盐水外,其余3组每日予50mg/kg雷公藤多苷片混悬液灌胃;针刺组每日予双侧"肾俞","关元"及"中脘"穴隔日一次轮流交替针刺,每5min行针10s,共留针10min。其余3组给予同步抓取。雌二醇组另予0.2mg/kg戊酸雌二醇片混悬液灌胃。连续干预14天。结果与空白组相比,模型组卵巢指数明显下降(P<0.01),血清E_2下降,FSH、LH上升(P<0.01);与模型组相比,针刺组和雌二醇组卵巢指数上升(P<0.01),血清E_2上升,FSH、LH下降(P<0.01)。HE染色见模型组卵泡及黄体减少,闭锁卵泡增多;针刺组及雌二醇组黄体、卵泡数量增多,闭锁卵泡数量减少。电镜下模型组颗粒细胞排列分散,细胞器损坏,脂滴减少;针刺组及雌二醇组细胞器损坏减少,颗粒细胞排列紧密,细胞器损坏较少,脂滴丰富。结论针刺可以保护雷公藤所致卵巢功能减退。Objective To observe the protective effect of acupuncture on ovary dysfunction of model mice induced by Tripterygium.Methods 40 normal estrus mice were randomly divided into 4 groups with 10 rats in each group.They were blank group,model group,estradiol group and acupuncture group.Except for the blank group gavaged with the same volume of normal saline was given per day,the other three groups were given daily 50 mg/kg glycosides of Tripterygium glycosides tablet;the acupuncture group was given bilateral"Shenshu"/"Guanyuan"&"Zhongwan"daily.And the acupuncture points alternately alternates acupuncture every other day,every 10 minutes for 10 seconds and a total of 10 minutes.The other 3 groups were given simultaneous capture.The estradiol group received 0.2 mg/kg estradiol valerate suspension.Continuous intervention for 14 days.Results Compared with the blank group,the ovarian index of the model group decreased significantly(P<0.01),serum E2 decreased,FSH and LH increased(P<0.01).Compared with the model group,the ovarian index increased in the acupuncture group and the estrogen group(P<0.01),serum E2 increased,FSH,and LH decreased(P<0.01).HE staining showed decreased follicles and corpus luteum and increased number of atresia follicles in the model group,increased number of corpus luteum and follicles in the acupuncture group and estradiol group,and decreased number of atresia follicles.Under the electron microscope,the granulosa cells of the model group were dispersed and the organelles were damaged,and the lipid droplets were reduced.The organelle damage was reduced in the acupuncture group and the estradiol group,the granular cells were arranged closely,the organelles were less damaged,and the lipid droplets were abundant.Conclusion Acupuncture can protect the ovarian function caused by Tripterygium wilfordii.
