摘  要:目的观察钛支架微孔参数(孔径、孔隙率)对成骨细胞黏附、增殖、分化的影响。方法根据钛支架微孔参数分A、B、C、D、E、F组。将小鼠胚胎成骨前体(MC3T3-E1)细胞接种于各组支架,扫描电子显微镜(SEM)观察细胞黏附;阿尔玛蓝(Alamarblue)法检测细胞增殖;实时定量聚合酶链反应(PCR)检测细胞碱性磷酸酶(ALP)、Runt相关转录因子2(Runx2)、1型胶原(Col-1)、骨钙素(OCN)表达。结果(1)细胞黏附:A、B组比C、D、E组细胞伪足多、黏附形态好。(2)细胞增殖:各组细胞相对增殖率差异有统计学意义(P<0.01),E组(1.729±0.180、2.148±0.275、2.680±0.175)高于其余各组(P<0.05)。(3)细胞成骨表达:各组细胞ALP、Runx2、Col-1、OCN表达差异有统计学意义(P<0.01)。A组(2.141±0.085、1.599±0.062、1.701±0.096,2.703±0.076)高于其余各组(P<0.05)。结论微孔参数(297.903 μm,58.623%)的钛支架利于细胞黏附分化,微孔参数(803.902 μm,58.162%)的钛支架利于细胞增殖。Objective To study effect of 3D printed titanium scaffold pore parameter on osteoblast. Methods Mouse embryonic osteoblast precursor (MC3T3-E1) cells were cultured on 5 groups of scaffolds. The adhesion, proliferation and differentiation were tested by scanning electron microscope (SEM), Alamarblue and real-time polymerase chain reaction (PCR). Results Morphology of cell adhesion in group A, B was better than that in group C, D, E. Revealed that relative proliferation rates of each group were statistically significant (P<0.01);relative proliferation rates in group E (1.729±0.180, 2.148±0.275, 2.680±0.175) were higher than those in other groups (P<0.05). Gene expressions of each group were statistically significant (P<0.01);gene expressions in group A (2.141±0.085, 1.599±0.062, 1.701±0.096, 2.703±0.076) were higher than those in other groups (P<0.05). Conclusion The titanium scaffold with mean pore parameter of 297.903 μm and 58.623% facilitates the adhesion and differentiation and of 803.902 μm and 58.162% facilitates the proliferation of osteoblast.
