摘  要:目的建立一种高效清除并重建肝巨噬细胞的方法,检测敲除ABRO1对肝巨噬细胞重建的影响。方法8周龄CD45. 1亚型C57BL/6J小鼠,尾静脉注射氯氟松(clophosome)清除肝巨噬细胞,钴源照射后,移植Abro1+/+或Abro1-/-小鼠(CD45. 2)骨髓,8周后分离小鼠肝非实质细胞,流式分析检测肝巨噬细胞的重建效率。结果常规骨髓移植后,肝脏中单核来源巨噬细胞(CD11bhiF4/80lo)重建率高达90%,但Kupffer细胞(CD11bintF4/80hi)的重建率只有约10%;提前注射氯氟松后再进行骨髓移植,肝脏中单核来源巨噬细胞和Kupffer细胞的重建率都>90%。利用氯氟松清除肝巨噬细胞后再进行骨髓移植的方法,检测发现敲除ABRO1并不影响肝巨噬细胞的重建。结论通过采用尾静脉注射氯氟松联合骨髓移植的方法,成功建立了一种安全、高效清除并重建肝内巨噬细胞的方法,敲除ABRO1并不影响肝巨噬细胞的重建。Objective To establish an efficient method for removing and reconstructing hepatic macrophages,and detect whether ABRO1 knockout affects the reconstruction of hepatic macrophages. Methods Eight-week-old CD45. 1+C57 BL/6 J mice were injected with clophosome to remove liver macrophages. After cobalt irradiation,bone marrow of Abro1+/+or Abro1-/-( CD45. 2) was transplanted into CD45. 1+mice. Subsequently,after 8 weeks of bone marrow transplantation,the clearance and reconstruction efficiency of hepatic macrophages were detected by flow cytometry. Results In the cobalt irradiation group,the removal and reconstruction efficiency of CD11 bhiF4/80 locells was up to 90%,but that of CD11 bintF4/80 hicells was only 10%. In clophosome scavenging and cobalt irradiation group, the removal and reconstruction efficiency of CD11 bhiF4/80 locells and CD11 bintF4/80 hicells was both up to 95%. Using clophosome scavenging plus cobalt irradiation methods,we found that ABRO1 knockout did not affect the reconstruction of hepatic macrophages. Conclusion We have established a safe and efficient method for reconstructing hepatic macrophages by using the combination of clophosome clearance and cobalt irradiation. ABRO1 knockout does not affect the reconstruction of hepatic macrophages.
