摘 要:目的探讨软坚散结方(RJSJ)对5-氟尿嘧啶(5-Fu)药动学参数的影响,并考察RJSJ或(和) 5-Fu对艾氏(EAC)荷瘤小鼠生命延长的作用。方法建立小鼠血清中5-Fu的HPLC-UV分析方法,将小鼠随机分为5-Fu组和RJSJ+5-Fu组,按时间点采集血样处理并计算5-Fu的药动学参数。小鼠腹腔接种EAC细胞,建立移植肿瘤模型。将动物分为对照组、5-Fu组、RJSJ组和RJSJ+5-Fu组,记录动物的存活情况,计算中位生存期(MST)和生命延长率(ILS)。结果与5-Fu单独给药组相比,联合用药组能显著增加体内平均滞留时间MRT(0-∞)(P <0. 05)。各给药组动物MST较模型对照组明显增加,5-Fu组、RJSJ组及RJSJ+5-Fu组动物ILS分别为31. 82%、34. 14%及45. 45%。结论 RJSJ可影响5-Fu在小鼠体内的药动学过程,二者单用或联用对小鼠EAC腹水瘤均有抑制作用。Objective To investigate the effects of Ruanjian Sanjie prescription( RJSJ) on 5-fluorouracil( 5-Fu pharmacokinetic parameters,and evaluate the influence of RJSJ and/or 5-Fu on life extension in Ehrlich ascites tumour( EAC) bearing mice. Methods An HPLC-UV method was established to determine 5-Fu in mouse serum. The mice were randomly divided into the 5-Fu alone group and combination with RJSJ group. Blood samples were collected at different time points and the pharmacokinetic parameters were measured. The transplanted tumor model of mice was established by intraperitoneal inoculation of EAC cells. The mice with EAC were divided into control,5-Fu,RJSJ and RJSJ + 5-Fu groups. Animal survival of each group was observed before the median survival time( MST) and the increase in life span( ILS) were calculated. Results Compared with 5-Fu alone group,MRT( 0-∞)of 5-Fu was significantly increased in the combination group( P < 0. 05). Compared with the model control group,MST in each experimental group was increased obviously. The ILS of 5-Fu group,RJSJ and RJSJ + 5-Fu groups was 31. 82%,34. 14% and 45. 45%,respectively.Conclusion The pharmacokinetics process of 5-Fu can be changed by RJSJ to some extent. RJSJ alone or in combination with 5-Fu can help inhibit EAC tumor in mice.
