摘  要:目的:观察背部循经推拿对亚健康模型大鼠海马神经元凋亡相关基因Bcl-2、Bax及Casepase-3蛋白表达的影响。方法:60只wistar大鼠随机分为正常组、模型组和推拿干预组,后两组予束缚应激,推拿干预组予背部循经推拿。实验结束后,采用免疫组化法检测3组大鼠海马组织Bcl-2、Bax及Casepase-3蛋白表达,实时荧光定量PCR方法检测Casepase-3 mRNA的表达情况。结果:模型组大鼠海马组织Bcl-2蛋白表达下降、Bax与Caspase-3蛋白表达上升,与空白组比较,差异有显著性(P<0.05);推拿干预组海马组织Bcl-2蛋白表达上调、Bax蛋白表达下调、Caspase-3蛋白及Caspase-3 mRNA表达均上升,与模型组比较,差异有显著性(P<0.05)。结论:背部循经推拿手法可以通过抑制Bax与Caspase-3蛋白及Caspase-3 mRNA表达、上调Bcl-2蛋白表达,从而发挥对慢性应激所致亚健康状态的脑神经保护作用。Objective: To observe the effect of Chinese massaging along the meridians on the back on expressions of apoptosis related genes Bel -2, Bax and Casepase - 3 in hippocampal neurons of sub - health model rats. Methods: Sixty Wister rats were divided into normal feeding group, model group and Chinese Tuina intervention group. The latter two groups were treated with restraint stress. Besides, the Tuina group used Tuina. Expressions of Bel - 2, Bax and Casepase - 3 protein in 3 groups of rats were detected by immunohistochemistiy. The expression of Casepase - 3 mRNA was detected by real - time quantitative PCR. Results: The expression of Bel - 2 protein in hippocampus of model rats decreased and the expressions of Bax and Caspase -3 protein increased. The difference was significant comparing with control group ( P < 0. 05 ). In the Tuina intervention group, expressions of Bel -2 protein, Caspase-3 protein and Caspase -3 mRNA increased and the expression of Bax protein decreased. Compared with the model group, the difference was significant (P < 0. 05). Conclusion : Chinese massaging along the meridians on the back can inhibit the expressions of Bax, Caspase-3 protein, Caspase-3 mRNA and upregulate the expression of Bel - 2 protein so as to exert the effect on chronic stress induced sub health state.
