慢性应激对卵巢早衰大鼠血清中E_2、FSH、LH及β- EP水平的影响

摘　　要：目的:通过检测慢性应激致卵巢早衰大鼠血清中E_2、FSH、LH、β-EP水平及观察卵巢组织形态以及激素水平的变化,探讨中药木尼孜其合剂对卵巢早衰的作用。方法:选用90只性成熟Wistar雌鼠,随机选取10只设为正常组(C组),其余80只为应激模型组,模型建成之后从中筛选出成卵巢早衰者分为POF模型组(POF组)及POF药物干预高、中、低剂量组,用ELISA试剂盒检测大鼠血清中E_2、FSH、LH、β-EP激素水平及观察卵巢组织形态变化。结果:木尼孜其合剂对慢性应激致卵巢早衰大鼠卵巢组织形态有一定的改善作用。与正常对照组比较模型对照组的E_2、β-EP激素水平明显降低(P<0.05), LH、FSH的水平显著升高(P<0.05);与模型对照组比较,各药物干预药组上述指标均有不同程度的改变(P<0.05)。结论:慢性应激(湿、寒、光照)能够导致POF的发生,中药木尼孜其合剂有防治卵巢早衰的作用,其作用机制与能够改变血清中E2、β-EP、LH及FSH等激素水平有关。Objective: To explore the effects of traditional Chinese medicine Muniziqi mixture on chronic stress premature ovarian failure by testing the serum levels of E2, FSH and β-EP in rats with chronic stress premature ovarian failure and observing the changes of ovarian tissue morphology and hormone levels. Methods: Ninety mature Wistar female rats were selected, 10 of them were taken randomly as control and the rest of them were used to make chronic stress model. After chronic stress model was established, rats which were diagnosed as POF disease group( POF), POF disease treated with high, middle and low doses of Chinese medicine groups. Result: Traditional Chinese medicine Muniziqi mixture worked on ovarian morphology of rats with chronic stress premature ovarian failure. Compared with the normal control group, the levels of E2 and β-EP in the control group were significantly lower(P < 0. 05 ), and the level of FSH was significantly higher( P <0. 05). Compared with the model control group, the above indicators of the drug intervention group had change in degree ( P < 0. 05 ). Conclusion: Chronic stress ( damp, cold, strong light) can lead to the occurrence of premature ovarian failure( POF). Traditional Chinese medicine Muniziqi mixture has the role of prevention and treatment of premature ovarian failure. The mechanism is related to the improvement of E2, FSH, LH and β-EP levels.
