乌灵胶囊联合卡马西平的抗癫痫作用以及对大鼠海马氨基酸含量的影响

摘　　要：目的:研究乌灵胶囊联合卡马西平(CBZ)对海人酸致癫痫大鼠痫性发作的干预作用,揭示联合使用乌灵胶囊可增强CBZ抗癫痫作用的机制。方法:通过颅内微量注射海人酸诱导癫痫大鼠模型,采用癫痫Racine等级对各给药组大鼠进行评分,以判断单用CBZ及联合给药的抗癫痫作用,采用HPLC-FLD法检测各组大鼠海马组织内Asp,Glu,Gly和GABA含量。结果:乌灵胶囊联合CBZ能够显著提高CBZ的抗癫痫作用,联合给药可使癫痫大鼠海马组织内Glu含量显著降低、Gly和GABA含量显著升高、Asp/Gly和Glu/GABA水平显著降低(P <0. 05)。结论:乌灵胶囊联合CBZ能够增强CBZ的抗癫痫作用,并能调节大鼠海马组织内兴奋性与抑制性氨基酸类神经递质的平衡。Objective: To study the effect of Wuling capsule combined with carbamazepine( CBZ) on seizures induced by kainate in rats,and to reveal the anti-epileptic mechanism of CBZ combined with Wuling capsule. Methods: The rat model of epilepsy induced by intracranial microinjection of kainite was used to score the rats in each group by epileptic Racine grade to judge the anti-epileptic effect of CBZ alone or in combination. The content of Asp,Glu,Gly and GABA in hippocampus of rats in each group was detected by HPLC-FLD method.Results: Wuling capsule combined with CBZ can significantly improve the anti-epileptic effect of CBZ. Wuling capsule combined with CBZ can significantly reduce Glu content,increase Gly and GABA content,and decrease Asp/Gly and Glu/GABA levels( P < 0. 05) in hippocampus of rats with epilepsy. Conclusion: Wuling capsule combined with CBZ can enhance the anti-epileptic effect of CBZ and regulate the balance of excitatory and inhibitory amino acid neurotransmitters in rat hippocampus.
