晋南1号枣树花药培养再生株系的染色体型组分析

摘　　要：染色体组型分析是细胞遗传学和遗传育种学研究的基础方法之一。染色体作为基因的载体,具有物种特异性,对研究物种分类、演化以及染色体结构、形态与功能之间的关系有非常重要的意义。以晋南1号枣树花药培养得到的第6代试管苗幼嫩叶片为材料,采用酶解去壁低渗-改良苯酚品红染色法制片、辅助显微观察分析,研究了其染色体组型。结果表明,该株系试管枣苗的染色体数目为12条,含有1个染色体组,属小染色体;其染色体组型公式为n=1X=12=9m+3sm,属2B型。在理论上为该枣树花药培养再生株系的快繁与育种利用提供了遗传种质依据;同时,提供了枣树染色体组分析的改进方法。The analysis of chromosome karyotype is one of the basic methods for the study of cytogenetics and genetic breeding.As a carrier of genes,chromosome has species-specific,which are of great significance for studying of the evolution classification of species and the relationship among the structures morphology and functions of chromosome.The tender leaves of the 6th generation of test tube seedlings obtained from anther culture of Zizyphus jujube mill Jinnan No.1 were selected as materials,the enzyme solution to wall low permeability-modified phenol fuchsin staining method was used to make specimen slice and auxiliary microscopic observation analysis was used to study its karyotype.The results showed that the number of chromosomes in this test tube seedlings were 12,including 1 chromosome group and the small chromosome.The formula of chromosome group is n=1X=12=9m+3sm,belonging to type 2B.In theory,this study would provide genetic basis for the rapid propagation and breeding of anther culture regeneration plant from this Chinese jujube.At the same time,the improved method for the analysis of jujube chromosome group was provided.
