摘 要:目的探讨微小RNA-7(miR-7)与肝细胞性肝癌(HCC)临床病理特征及上皮间质化转变(EMT)之间的相关性。方法收集30组HCC(包括癌组织及对应癌旁组织)临床标本及临床相关病理资料,分别检测miR-7及EMT标志物N-钙粘蛋白(N-cadherin)、E-钙粘蛋白(E-cadherin)的表达,分析miR-7与HCC临床病理特征之间的相关性及miR-7与EMT之间的相关性。结果 miR-7、E-cadherin在癌组织的表达水平显著低于癌旁组织(P<0.01),而N-cadherin的表达则相反。相关性分析表明,miR-7与E-cadherin呈正相关性,与N-cadherin呈负相关性。miR-7在HCC的表达与患者年龄、性别、肿瘤部位、是否伴随肝炎病毒感染无相关性,而与TNM分级、分化程度和血管侵犯密切相关。结论 miR-7的表达水平与EMT程度呈负相关性,检测miR-7的表达对HCC的临床诊断、治疗及预后判断具有潜在的重要意义。Objective To investigate the relationship between miR-7 and clinicopathological features of hepatocellular carcinoma and hepatocellular carcinoma(EMT)in epithelial mesenchymal transition(HCC).Methods Real-time PCR was used to detect the expression of miR-7 in 30 HCC cancer tissues and paracancer tissues.Real-time PCR and Western blot was applied to detect the mRNA and protein expression of N-cadherin and E-cadherin.Meanwhile,the correlation between miR-7 and clinicopathological features of hepatocellular carcinoma and EMT was analyzed by SPSS 19.0.Results The expression levels of miR-7 and E-cadherin in cancer tissues were significantly lower than those in paracancer tissues(P<0.01).However,the expression of N-cadherin was significantly increased(P<0.01).Correlation analysis showed that miR-7 was positively correlated with E-cadherin and negatively correlated with N-cadherin.Meanwhile,the expression of miR-7 in HCC was not associated with patient age,gender,tumor location,or hepatitis virus infection,but was closely related to TNM grade,degree of differentiation and vascular invasion.Conclusion The expression level of miR-7 is closely related to HCC EMT.Detecting the expression of miR-7 has potential significance in clinical diagnosis,treatment and prognosis of HCC.
