摘 要:目的研究龙血竭总黄酮对猪血清诱导的大鼠免疫性肝纤维化的影响及其可能的分子学机制。方法实验通过建立免疫性肝纤维化大鼠模型模拟临床上由病毒性肝炎引起的肝纤维化疾病,大鼠造模第16周后处死,分别取肝脏左叶固定并进行HE染色,显微镜下观察肝纤维化程度;采用ELISA法检测大鼠血清中透明质酸(HA)、Ⅲ型前胶原(PCⅢ)、层粘连蛋白(LN)和Ⅳ型胶原(CⅣ)的水平;同时用ELISA法测定肝脏组织中环磷腺苷(cAMP)的含量;Western blot法定量分析大鼠肝脏组织中转化生长因子β_1(TGF-β_1)蛋白表达情况。结果与正常对照组比较,模型组大鼠血清中HA、PCⅢ、LN及CⅣ的含量明显升高(P<0.01),肝组织匀浆中cAMP水平明显下降(P<0.01),TGF-β_1蛋白表达显著升高(P<0.01),同时肝脏中肝小叶结构被破坏,胶原分泌增加;与模型组比较,龙血竭总黄酮高、中剂量组大鼠血清中HA、PCⅢ、LN及CⅣ的含量明显降低(P<0.05),肝组织匀浆中cAMP水平明显升高(P<0.01),TGF-β_1蛋白表达显著降低(P<0.01),并且肝纤维化程度减轻。结论龙血竭总黄酮可能通过降低肝脏内TGF-β_1蛋白的表达抑制肝星状细胞的增殖和细胞外基质的合成及降解,达到减轻肝纤维化的目的。Objective To investigate the effect of Sanguis Draconis flavones(SDF)in immunologic hepatic fibrosis(IHF)induced by pig serum in rats and its possible mechanism.Methods The immune liver fibrosis rat model was established to simulate the clinical hepatic fibrosis caused by viral hepatitis,the left lobe of the liver was fixed and stained with HE respectively after 16 weeks of IHF rats,and the degree of liver fibrosis was observed by microscope.The level of HA,PCⅢ,LN and CⅣin serum and the content of cAMP in liver tissue homogenate were detected by ELISA,and the expression of TGF-β1 protein in liver tissue of rats was determined as well.Results Compared with normal group,the contents of HA,PCⅢ,LN and CⅣin serum in model group rats were increased significantly(P<0.01),the level of cAMP in liver tissue homogenate significantly decreased(P<0.01),the TGF-β1 protein expression was significantly increased(P<0.01),at the same time,the structure of hepatic lobule was damaged in the liver and the collagen secretion increased.Compared with model group,the contents of HA,PCⅢ,LN and CⅣin rat serum in SDF high dose group and middle dose group significantly decreased(P<0.05),the level of cAMP in liver tissue homogenate increased significantly(P<0.01),the TGF-β1 protein expression significantly reduced(P<0.01),and the degree of liver fibrosis decreased.Conclusion SDF can reduce liver fibrosis,and its mechanism may be related to reducing the expression of TGF-β1 protein in the liver,inhibiting the proliferation of HSC and the synthesis and degradation of ECM.
