摘 要:目的:淡黄花百合中多糖含量的研究目前未见文献报道,优选最佳条件,建立淡黄花百合中多糖的含量测定方法。方法:以葡萄糖为标准品,采用苯酚-硫酸法测定。结果:确定了检测波长为489.5 nm,显色条件为常温下避光静置40 min,最佳方法为回流提取法,沸腾后回流1.5 h;测得12个批次贵州不同产地的淡黄花百合中多糖的百分含量范围为18.58%~37.53%,平均含量为24.07%。结论:本实验建立的淡黄花百合多糖含量测定方法简便、准确、易操作、实用性强。不同产地的淡黄花百合中多糖的含量存在差异,实验结果为山百合药材质量研究提供新的依据。Objective: To explore the best extraction conditions for the determination of polysaccharides in Lilium sulphureum Baker of Guizhou Province. Methods: Glucose was used as a standard and the phenol-sulfuric acid method was applied for the dertemination. Results: The detection wave length was 489.5 nm;the optimal time for coloration was 40 minutes at room temperature in the dark;the best extraction condition was reflux extraction method, and reflux time was 1.5h after boiling;the percentage of polysaccharide in the 12 batches of Lilium sulphureum Baker of different production areas in Guizhou ranged from 18.58% to 37.53% with an average content of 24.07%. Conclusion: The method for determining the polysaccharide content of Lilium sulphureum Baker established in this experiment is practical, and it is easy to operate and accurate. The difference of polysaccharide content in Lilium sulphureum Baker of different habitats could provide a new basis for the study of the quality of Bulbus lilii.
