摘  要:目的探讨解脲脲原体(Uu)感染对男性不育患者精子血管内皮生长因子(VEGF)及其受体(VEGFR2)表达和精子顶体酶活性的影响。方法参照世界卫生组织(WHO)标准方法对175例精液标本进行精液常规分析,其中正常生育组30例,不育男性145例。Western blot检测VEGF蛋白在人精子中的表达,免疫荧光观察VEGFR2在人精子上的定位,采用改良明胶底物膜法检测精子顶体内顶体酶(arcosin)活性(arcosin阳性反应率、arcosin活性强度)。结果 145例男性不育症患者中精液Uu阳性47例,阳性率32.41%(47/145)。Uu阳性不育组的精子VEGF/GAPDH平均光密度均明显低于Uu阴性不育组和正常生育组(P <0.05)。Uu阳性不育组的精子VEGFR2阳性率明显低于Uu阴性不育组和正常生育组,经统计学分析差异具有统计学意义(P <0.05)。Uu阳性不育组的arcosin活性(arcosin阳性反应率、arcosin活性强度)明显低于Uu阴性不育组和正常生育组(P <0.05)。相关性分析提示精子VEGF蛋白表达量与arcosin活性存在正相关性;精子VEGFR2阳性率与arcosin活性存在正相关性。结论 Uu感染阳性不育患者精子VEGF蛋白及其受体VEGFR2表达降低伴随arcosin活性(arcosin阳性反应率、arcosin活性强度)减弱。Objective To study the effect of Ureaplasma urealyticum(Uu)on expression of vascular endo thelial growth factor(V EGF) and its receptor( VEGFR2) and activity of acrosin in human spermatozoa. Methods 175 cases of semen samples were collected. 145 cases were in infertile group,and 30 cases in normal control group. Semen routine analysis was referred to the WHO standard. Western blotting was used to detect the level of VEGF expression in spermatozoa. The localization of VEGFR2 protein on human spermatozoa was determined by indirect immunofluorescent staining. The acrosin activity(acrosin positive rate,acrosin activity intensity) in all samples were examined by improved fixed substrate film method. Results The expression of VEGF protein and the percentage of the VEGFR2 positive rate in Uu positive culture infertile group were lower than those in Uu negative culture infertile group and control group with statistical difference(P < 0.05). The activity of acrosin( acrosin posi tive rate,acrosin activity intensity) in Uu positive culture infertile group were lower than those in Uu negative culture infertile group and control group with statistical difference. A significant positive correlation was found among the VEGF protein expression and VEGFR2 positive rate with acrosin positive rate and acrosin activity intensity. Conclusion Ureaplasma urealyticum infection may affect the expression of VEGF protein,the positive rate of VEGFR2 and activity of acrosin( acrosin positive rate,acrosin activity intensity) in human spermatozoa.
